| Inner Mongolia cashmere goat semen liquid preservation is one of the main means of genetic resource preservation,which can effectively improve the utilization efficiency of germplasm resources.The cryopreservation of semen is mainly to reduce or inhibit the active metabolism of sperm to prolong the survival time in vitro.Different antioxidants have different effects on the liquid preservation of semen.In this experiment,the effects of different cooling rates on the cryopreservation of semen were compared.On this basis,the effects of different concentrations of astaxanthin and melatonin on the semen preservation of Inner Mongolia cashmere goats were studied.The test results are as follows:1.Different cooling rates have different effects on fresh sperm diluent.When the cooling rate is controlled to 7.75℃/h,the sperm motility is 81.9%,which is significantly higher than other cooling time groups(P<0.05).When the cooling rate was controlled at 15.5,7.75,5.17,and 3.88℃/h,there was no significant difference in the sperm motility of fast forward movement(P>0.05).The changes of sperm-related motion parameters VAP,VSL,and VCL values can further illustrate that the cooling time control at 4h is more suitable for production practice.2.Adding antioxidants in the liquid storage of semen can effectively improve the quality of sperm.Adding 2 μM astaxanthin can keep the semen stored for 7 days,and the sperm motility is more than 70%,which is significantly higher than that of the control group(P<0.05).The concentrations of 0.5,1,and 4 μM astaxanthin were higher than the control group,but there was no significant difference(P>0.05).Astaxanthin had little effect on sperm-related motor parameters,but the value of the 2μM astaxanthin group was slightly higher than that of the control group.Therefore,the addition of 2 μM astaxanthin in the diluent can improve sperm motility.3.Adding 0.1 mM melatonin to the sperm dilution of Inner Mongolian cashmere goats can significantly improve sperm motility(P<0.05),and its related exercise parameters also increase accordingly.In summary,during the cryopreservation process of the Inner Mongolian cashmere goat semen,controlling the cooling rate to 4 hours can further improve the effect of sperm preservation.On this basis,adding an appropriate amount of astaxanthin or mela-tonin can better protect sperm. |
