Lycium ruthenicum Murr.is one of the native plants.With the deepening research from different directions,the importance of its medicinal value,economic value and ecological value has been gradually emerged.In order to rationally use and protect the fruit of L.ruthenicum Murr.,this research works from two aspects: one is the metabolites polyphenols that represent the quality of L.ruthenicum Murr,the other is characteristics of soil salinity in the native place.The purpose is to grasp the response relationship between polyphenols and soil salinity,guide the scale selection of planting site conditions and promote the development of the plateau-specific berry planting industry.NaNO2-Al?NO3?3-Na OH colorimetric method was used to determine total flavonoids,and High-performance Liquid Chromatography-mass Spectrometry?HPLC-MS/MS?was used to identify p-anthocyanins.Combined with Ultra-high Performance Lliquid Chromatography-tandem Quadrupole Mass Spectrometry?UPLC-Q-Orbitrap MS?technology to analyze polyphenol components,drying methods,flame photometry and other classic methods are used to analyze and determine the soil salinity.Finally,through appropriate mathematical and statistical processing,the response relationship of polyphenols from L.ruthenicum Murr.to soil nutrients was explored.The total flavonoid content in L.ruthenicum Murr.of Qaidam was 1.63±0.10%?3.15±0.16%,and the total polyphenol content was 8.19±0.50?28.60±0.54mg·g-1.A total of 12 anthocyanins were isolated and identified.Petunia-3-O-cis?or trans?coumarinylrutin-5-O-glucoside is the main component,accounting for 49.42%?79.13% of the total anthocyanins.Followed by petunidin-3-O-p-coumaric acid-sophoroside-5-O-glucoside is accounting for 2.28%?18.22% of total anthocyanins;caffeic acid,Kaempferol,naringenin,p-coumaric acid,quercetin and rutin are the main non-anthocyanin polyphenol components.The main native soil in L.ruthenicum Murr.of Qaidam is alkaline chloride-type saline soil,and the soluble salts are mainly Na Cl,followed by KCl and Ca Cl2.The soil salt content has a distribution pattern of southern content higher than northern and western content higher than eastern.There is a positive correlation between soil salt content and transpiration ratio;Mg2+ and CO32-have strong variability,HCO3-,K+,SO42-,Na+,Ca2+,and Cl- have moderate variability in order of increasing order,conductivity and soluble salt The content has a medium degree of variation,and the p H is weak.The quadratic fitting relationship between the soil soluble salt content and the conductivity of the 1:1 extract solution has the best fitting effect?R2=0.9755?.The optimal function model can be used to quickly determine the soil soluble salt content by the conductivity method.The formation of polyphenols in L.ruthenicum Murr.of Qaidam is affected by the combination of climatic factors and soil salinity.Cold-temperature cycling arid environment is favorable for polyphenol production,and the total polyphenol content is weakly negatively related to the soil soluble salt content.Too high soil soluble salt content will inhibit the synthesis of naringenin;The content of quercetin,rutin,and total flavones is poorly correlated with climate,but is significantly positively correlated with CO32-in the soil.K+ can promote the synthesis and accumulation of flavonoids in L.ruthenicum Murr.Ca2+ may be the main ion that negatively affects the total polyphenols in L.ruthenicum Murr.
