This paper through 3 wild strains of Pleurotus ostreatus were identified by morphology,antagonistic test,determination of esterase Isozyme,identification of ISSR molecular markers,Its identification and phylogenetic tree was established to determine the species relationship among the 3 strains,in order to construct the information database of wild Pleurotus ostreatus strains and enrich Pleurotus ostreatus germplasm resources in China and play a certain role.The biological characteristics of 3 wild strains of Pleurotus ostreatus were evaluated,high quality Pleurotus ostreatus germplasm was screened out,it provides abundant materials for cross breeding of Pleurotus ostreatus.The results are as follows.1.Morphological identification of 3 wild strains of Pleurotus ostreatus and the known reference strains was carried out,3 wild strains of Pleurotus ostreatus were identified as different in morphology,there are certain differences between the strain and the production reference strain.2.3 strains of wild Pleurotus ostreatus and 3 kinds of reference strains were identified by antagonistic,esterase isozyme,ISSR and ITS sequence analysis,the results of antagonistic assay showed that the antagonistic effects of the two drugs were similar.There were obvious antagonistic reactions between the 3 wild strains and the 3 reference strains,and all of them belonged to the uplift type antagonistic type.The results of esterase isozyme showed that,a total of 49 esterase isozyme bands were detected in 6 strains,there were 15 polymorphic loci,it shows that the polymorphism is good.The isozyme patterns of ISSR and isozyme were analyzed,the3 wild strains of Pleurotus ostreatus were not the 3 known reference strains,and the 3wild strains did not belong to the same species.Its sequencing and phylogenetic tree of 3 wild strains showed that,PO14belongs to Pleurotus sapidus,PO15belongs to Pleurotus ostreatus and PO37belongs to Pleurotus ostreatus.3.The biological characteristics of 3 wild strains of Pleurotus ostreatus were evaluated,the mycelial culture of 3 wild original species,fruiting,The biological conversion rate test showed that strain PO15is a potential high yield and high quality strain.When the ambient temperature is between 15?to 30?,the growth rate of 3wild strains was faster than that of PO2.At 35?,PO14did not germinate.The optimum nutrient water content of wild strains PO14and PO37was 55%,wild strain PO15and production reference strain PO2,the optimum nutrient water content was 60%.At the same p H value of the medium,there was no significant difference in the growth rate of different strains,the growth rate of the same strain was significantly different in different p H medium.The evaluation of resistance of rhizopus showed that,the hyphae of the 3 wild strains showed obvious resistance to rhizopus.
