Rhizoma Coptis, as a native herb to Chongqing. With reference to the2010edition of Pharmacopoeia, for a better development and utilization of it’s medicinal value. This paper aims to make a research on coptis’s accessory substance. The contents and results are as follows:1. The different duration of rhizoma coptidis alkaloid content in the plant and determined the rhizoma coptidis in different regionsPillars of take two years, three years, four years, five years of rhizoma coptidis plants, Removed must, stem, leaf and rhizoma coptidis fibrous under60℃oven, after sample drying combined with HPLC fingerprint determination of the coptis chinensis leaf, rhizoma coptidis, rhizoma coptidis alkaloid content in the hair root, the results showed that rhizoma coptidis plant at the time of the second year of growth to root rhizoma coptidis alkaloid, began to accumulate to fifth years rhizome alkaloid increased to11.158%, and rhizoma coptidis shall and coptis chinensis leaf alkaloid from3.965%,2.136%decreased to2.798%,1.376%, that rhizoma coptidis alkaloids as growth continues to shift to the roots of plants.From six different origin of rhizoma coptidis, rhizoma coptidis alkaloid content of them are analyzed, the results showed that September Yuelai rhizoma coptidis alkaloid content is higher in must, total alkaloid content of2.620%;In October Huangshui rhizoma coptidis alkaloid content in the highest,2.764%;In November made in Lichuan rhizoma coptidis alkaloid content in the highest,2.764%.So this can choose rhizoma coptidis must be of high quality production.Selection of rhizoma coptidis high-quality production areas, Huangshui’s rhizoma coptidis5years be measured respectively in each month of the year alkaloid content. To explore the same origin of rhizoma coptidis alkaloid content distribution in different months. The results showed that in January, the total alkaloids in rhizoma coptidis was0.887%, the cumulant maximum when October,2.620%;And then in November and December rhizoma coptidis alkaloid, for the best gathering in rhizoma coptidis including region should for October.2.Measuring alkaloid content in by-products from coptis production.With reference to the2010edition of Pharmacopoeia, we established the method of measuring the coptis’s alkaloids based on HPLC, combined with HPLC fingerprint, to measure the alkaloid content. We figured out the total alkaloid content of each part and optimized the use of the materials. The results showed that the alkaloid content of coptis leaves is low while the content of petioles and rhizoma coptidis is much higher. Petioles and rhizoma coptidis of coptis have higher medicinal value than coptis leaves. So petioles and rhizoma coptidis of coptis could be used as research subjects for comprehensive utilization.3.Experiment on the extraction process of coptis alkaloids.Using single factor experiment method, we experimented on the extraction acidity, solid-liquid ratio and extraction times separately, and analyzed and selected the optimal extraction conditions. The final parameters for extraction are:1.5%for H2SO4concentration,1:15for solid-liquid ratio,1h for extraction time.4.Preparation of coptis’s alkaloids monomer.This experiment adopted the method of precipitation agent and gradient elution. By concentrating the extract to100ml, after the addition of5%sodium chloride and20%of Na2SO4was recrystallized, we got berberine and coptisine. They were then combined with mother liquor on polyamide column, eluted with aqueous ammonia gradient lmol/L, and with the help of TLC tracking, decompression concentration after recrystallization, we got berberine and jatrorrhizine...
