acupuncture points(Guanyuan,Zhongji,Zigong,Sanyinjiao).By examining the OGTT and serum lipid metabolism indices(HLD-C,LDL-C,TG,TC)in each group of rats,we investigated whether low-frequency electroacupuncture can improve abnormal glucose and lipid metabolism in rats with PCOS and high-fat diet.Detection of liver oil red "O" staining and RT-PCR detection of liver tissue in each group(GSK3-β,INSR,GLUT1,GLUT4,PPARa,PPARy,SREBF1,SREBF2)mRNA content,to explore low-frequency electro-acupuncture Can improve the metabolic abnormalities(PCOS and obesity)model rat liver tissue sugar,lipid metabolism related factors,clarify that low-frequency electricity for PCOS patients and obese women with glucose and lipid metabolism disorders have a regulatory role and then play a therapeutic role.Methods:21-day-old clean female rats(SD)(body weight 50±5g)were selected,and then 48 female SD rats were randomly divided into 6 groups(n=8)by orientation.The blank group,blank acupuncture group,PCOS group,PCOS acupuncture group,high-fat diet group,high-fat diet acupuncture group;model:1 After randomized PCOS group,PCOS acupuncture group group began to give Rats were subcutaneously injected into the neck of the rats(DHEA(60mg/100g)was dissolved in 0.2 ml of soybean oil for injection),and the rest of the rats were injected with 0.2 ml of soybean oil for injection cycle of the rats in each group was observed;3 The HE staining of the ovaries of the rats.Low-frequency electro-acupuncture intervention:After evaluation of the model(21 days after modeling),low-frequency electro-acupuncture intervention was performed with continuous waves at a frequency of 2 Hz,1 time/day,5 days/week,and 5 consecutive weeks.15 minutes/time in the first week,20 minutes/time in the second and third weeks,and 25 minutes/time in the fourth and fifth weeks.The points are Guanyuan,Zhongji,Zigong,Sanyinjiao.Materials:The vaginal smears were taken 2 weeks before the end of the low-frequency electroacupuncture intervention.The estrus cycle of the rats in each group was observed.After entering the second regular estrous cycle,the eyelids were taken to the neck and sacrificed,and the rats were quickly taken out.Rat liver tissue was weighed and rapidly placed in liquid nitrogen and frozen and transferred to a-70℃ freezer for preservation.Detection index:OGTT,serum lipid metabolism related indicators,ovarian HE staining,liver tissue oil red"O" staining and RT-PCR detection of liver tissue in each group of rats(GSK-3β,INSR,GLUT1,GLUT4,PPARα,PPARγ acupuncture group and PCOS acupuncture were visible after acupuncture treatment.The body weights of rats in the acupuncture group and high-fat diet group were significantly lower than those in the blank acupuncture group,PCOS acupuncture group,and high-fat diet acupuncture group.Therefore,after the intervention of low-frequency electroacupuncture,there were The effect of weight loss in rats.2.PCOS rat model evaluation:In this experiment,the model group rats showed estrus cycle disorder on the estrous cycle.PCOS group compared with the blank group,PCOS acupuncture group and PCOS statistically significant difference between the PCOS acupuncture group and the PCOS group(p<0.05).4.Low-frequency electroacupuncture was used to study the metabolism of glucose and lipid in the liver tissue of each group:The mRNA expression of GSK3-β,INSR,GLUT1,GLUT4,PPARa,PPARy,SREBF1,and SREBF2 in liver tissue.Compared with PCOS group,GSK3-β blank group had statistically significant difference between PCOS acupuncture group and PCOS group(p<0.05).In the INSR blank group and PCOS group,PCOS acupuncture group had higher fat than PCOS group.Dietary acupuncture group was statistically significant compared with high-fat diet group(p<0.05);GLUT1,GLUT4 blank group compared with PCOS group,PCOS acupuncture group compared with PCOS group,high-fat diet acupuncture group and high-fat electroacupuncture can improve the treatment of polycystic ovary syndrome by improving the abnormality of liver and glucose metabolism in rats with polycystic ovary syndrome.3.Based on the results of this study,it is presumed that low-frequency electroacupuncture may improve the reproductive function of patients with polycystic ovary syndrome by improving liver lipid metabolism related factors in
