Objectives To explore the effects of Schisandra chinensis on myocardium in rats with hyperthyroid heart disease by Velocity Vector Imaging Technology(VVWI)and the protective mechanism of Schisandra chinensis on model rat heart.Methods Twenty male Sprague-Dawley(SD)rats aged 5-6 weeks were injected subcutaneously with L-thyroxin 0.5mg/kg for 28 days.Preparation of rat model of hyperthyroid heart disease.Divided into hyperthyroidism model group(HHD group,n=10)and Schisandra chinensis drug intervention group(SC group,n=10).In the SC group,the water extract of Schisandra chinensis was given 2 g/kg/d for 28 days.Control group(CON group,n=10).Subcutaneous injection of an equal volume of saline.At the same time,the same volume of saline was given to the stomach for 28 days.On the 29th day of the experiment,ultrasound velocity vector imaging technique was used to analyze myocardial motion in each group of rats.The levels of serum TT3?TT4 and HSP-70?NT-proBNP,the expression of P53 and activity of CaM,HE,Masson staining and electron microscopy in the treatment of hyperthyroid heart disease rats.Serum TT3,TT4,HSP-70 and NT-proBNP levels were detected by ELISA.P53 expression and CaM activity in heart tissue were detected by Western blotting.Pathological changes and ultrastructural changes of myocardium were observed by HE,Masson staining and electron microscopy.Results1.The systolic and diastolic radial velocity(Vs,Vd),radial strain(Sr),systolic and diastolic radial strain rate(SRr)of HHD group was lower than CON group(all P<0.05).The Vs?Vd?Sr and SRr of the SC group was higher than the HHD group(all P<0.05).2.The serum TT3?TT4 of HHD group was increased(all P<0.05)compared with the CON group,and was higher than the SC group(all P<0.05).The concentrations of HSP-70 and NT-proBNP in the HHD group and the SC group were higher than those in the CON group(all P<0.05),but there was no significant difference between the HHD group and the SC group(all P>0.05).3.The expression of P53 protein and CaM protein in the HHD group was increased,compared with the CON group.Compared with the HHD group,the P53 expression and CaM protein activity in the SC group decreased.4.Myocardial structure of the CON group showed no obvious pathological changes.Myocardial mitochondria in the HHD group showed marked swelling and interstitial fibrosis.Myocardial mitochondrial swelling in the SC group was reduced and interstitial fibrosis was reduced.Conclusion1.Schisandra chinensis has a protective effect on myocardial damage in rats with hyperthyroid heart disease,and its mechanism may be related to the down-regulation of P53 expression and CaM activity in Schisandra chinensis.2.?? can evaluate the myocardial movement in rats with hyperthyroid heart disease.3.Hyperthyroidism induces overexpression of HSP-70.
