Objective:By observing the effects of acupuncture at"Dazhui" and"Renzhong","Baihui" on neurological deficit score,cerebral infarction area ratio,apoptosis and mitochondrial ultrastructure in rats after cerebral ischemia-reperfusion,and to explore the effect of acupuncture on cerebral ischemia Mechanism of perfusion injury.Methods:A total of 96 96 male SD rats were randomly selected,24 of them were sham operated group,and the rest 72 rats were reconstructed by middle cerebral artery ischemia model(MCAO)with reference to Zea Longa line.After modeling,they were randomly divided into model group,acupuncture control point group,acupuncture point group,Each group has 24 rats,and then randomly divided into 24h group,72h group,two sub groups,Each group has 12rats.Model group,sham operation group only tied and do not acupuncture,acupuncture group acupuncture "Dazhui" and"Renzhong","Baihui" three acupoints,acupuncture control group acupuncture side next to the open at 0.3cm,Once every 12h,24h and 72h after treatment,the first rat nerve function scores after.Selected rats of ischemic brain tissue,detection of infarction area ratio,apoptotic cells were detected by transmission electron microscope,TUNEL method was used to observe the mitochondrial ultrastructure in the hippocampus of the ischemic cerebral TTC staining.Results:1.Comparison of neurological deficit score:72h after treatment,compared with sham operation group,model group of neural function defect score were increased,the difference was statistically significant(P<0.01);compared with the model group,acupuncture control group,acupuncturegroup of neurological deficit score were decreased,the difference was statistically(P<0.01orP<0.05);with the acupuncture point group,there were significant differences in neural function defect of acupuncture acupoint group(P>0.05),but the acupuncture group of neurological deficit score overall downward trend.Compared with the 24h group,the scores of nerve function defect in each group of the 72h groups were decreased.2.Comparison of the area ratio of cerebral infarction:24h after treatment,compared with sham operation group,the proportion of cerebral infarction area increased,the difference was statistically significant(P<0.01),and there was no significant difference in the ratio of cerebral infarction area between each group(P>0.05).72h after treatment,compared with sham operation group,the model of cerebral infarction area ratio increased,the difference was statistically significant(P<0.01);compared with the model group,the treatment group the cerebral infarction area ratio decreased(P<0.01),there was no significant difference in the treatment group(P>0.05).3.Comparison of the positive rate of apoptosis:72 h after treatment,compared with the model group,the positive rate of apoptotic cells in the acupuncture control point group and the acupuncture point group decreased(P<0.01 or P<0.05).Compared with 24 h,the positive rate of apoptotic cells in all 72h models decreased,but there was no significant difference in the model group(P>0.05).The difference between the acupuncture control group and the acupuncture point group was statistically significant(P<0.01 or P<0.05).4.Electron microscope observation showed that the sham operation group,model group and normal structure of mitochondria;mitochondrial swelling,cristae,cavity structure is obvious;the acupuncture control group showed mitochondrial swelling,cristae,small vacuoles;acupuncture group,mitochondria swelling,cristae and vacuolization is not obvious.Compared with the 24h group,72h group of mitochondrial structure and no changes in sham operated group,model group,mitochondrial ultrastructure was roughly,acupuncture group and acupuncture group were improved,the acupuncture control group cell edema and vacuolization degree were reduced,acupuncture group of mitochondrial structure has been relatively clear.Conclusion:1.Acupuncture in rats with "Dazhui" and "Renzhong","Baihui" three points can reduce cerebral ischemia reperfusion neurological function in rats,reduce the positive rate of cell apoptosis ratio and area cerebral infarction,improve mitochondrial ultrastructural changes,so as to play a protective effect on brain tissue.ameliorate the symptoms of neurological deficit in rats with cerebral ischemia and reperfusion,reduce the infarction area ratio and the positive rate of apoptosis,mitigate the damage of mitochondrial ultrastructure,Protection of brain tissue.2.The protective effect of acupuncture on brain tissue may be related to the improvement of mitochondrial ultrastructural damage by acupuncture,maintaining the integrity of mitochondrial structure and function,thereby inhibiting cell apoptosis.3.The protective effect of acupuncture on brain tissue is related to the length of acupuncture intervention,and the effect of 72h is better than that of 24h,In a certain period of time,there is a positive correlation between the therapeutic effect and the time of treatment.
