Objective:To study the effect of the Stimulation with Acupuncture (SA) on the injury repairing of Ischemic Brain Tissues. By observing the changes of related signal protein phosphorylation under SA, the related signal molecule and pathway were revealed, and from the proteome analysis, the mechanism of injury repairing of Ischemic Brain Tissues was expounded which riched the theory of meridian related with viscera, applied certain experimental and theoretic basis for clinical applying and was helpful to understand the therapeutic mechanism of Acupuncture.Methods:40 SD rats were randomly divided into 4 groups:sham-operat ed group, model group, contrast group and treatment group with 10 rats per group. The research was divided into two parts:part 1:The middle cerebral artery occlusion(MCAO) model was established by using the improveed thread embolism method. The scores of neurological functions deficits were evaluated after waking following operation and before sacrifice. The infarct volume in 10 rats of each group were studied by TTC staining after sacrifice. Part 2:The Ischemic Brain Tissues of the rats per group was extracted. The changes of related signal protein phos-phorylation of Ischemic Brain Tissues injury repairing with Acupuncture were detected by 720 kinds of phosphorylation of antibody protein chip tech. And the differential expressed phosphorylation signal protein in eac-h group was screened, then further revealed the related pathway to which the differential expressed phosphorylation signal protein belong.Results:1. Compared with sham group, the scores of nerve function defect and the percentage of brain infarct area of model group were increased significantly (P<0.01). however, Compared with model group and sham group, the scores of neurological functions deficits of treatment group were decreased in treatment and contrast group especially in treatment group(P<0.01 vs P<0.05),the difference between them was significant (P<0.05);2.720 kinds of phosphorylation of antibody protein chip tech detection shows:compared with sham group,48 kinds of protein were up-regulated more than 1.5 folds in the phosphorylation change,29 kinds were down-regulated in model group; compared with model group,51 kinds of protein were down-regulated more than 1.5 folds in the phosphorylation change,29 kinds were up-regulated in treatment group; while in contrast group,24 kinds were down-regulated and 35 were up-regulated; To conclude, the number, function type and signal transduction pathway of phosphorylation signal protein in the treatment group were more than that of contrast group.Conclusion:1. The neurological functions deficits in treatment group can be obviously improved and the Infarct volumes of treatment group was also lower then other groups, which indicates that Acupuncturing DA ZHUI, BAI HUI,REN ZHONG (ADBR) can obviously repair the Ischemic Brain Tissues,Besides, decreasing the size of infarction area is an important link of repairing the Ischemic Brain Tissues;2. ADBR can not only adjust the disordered level of protein phosphorylation that associated with the proliferation after making model, but also adjust those apoptosis-related disordered level of protein phosphorylation,and can regulate the proteins of model group which involves various functional types and belongs to multiple signal transduction pathways which include the cell cycle regulation of signal transduction pathway, MAPK signaling pathways, apoptosis signaling pathways, adhesion molecule signaling pathways, non-receptor tyrosine kinase signaling pathways, receptor tyrosine kinase signaling pathways such as signal transduction pathways. It indicates multi-target and multiple-way is the important characteristic of ADBR to repair the Ischemic Brain Tissues.3. Compared with Acupuncture contrast group, ADBS can activate the MAPK signal transduction pathway specifically which indicates MAPK signal transduction pathway is close related with repairing of. Acupuncture treating group to promote the repairing of Ischemic Brain Tissues.4. ADBR can obviously activate various functional types of proteins and several signal transduction pathway which accord with the basic characteristics of acupuncture such as holism and comprehensibility. Studying the holistic regulating effect of TCM and acupuncture by using protein chip tech can promote the understanding and applying of holism in TCM.
