Objective To detect the protein expression of SEMA3 A,SEMA3B,p53 and Ki-67 by immunohistochemical.Which discuss its relationship with clinical pathological parameters in mucoepidermoid carcinoma and the impact on clinic prognosis.Methods Collecting 71 cases of paraffin-embedded tissue samples in mucoepidermoid carcinoma and 35 cases in normal salivary glands.Detecting the protein level expression of SEMA3 A,SEMA3B,p53 and Ki-67 by immunohistochemical Envision two-step method in mucoepidermoid carcinoma and normal salivary glands.SPSS 20.0 statistical software were used to analyze the relationship between the related protein expression and the clinicopathological parameters.Kaplan-meier and Cox proportional risk regression models were used to analyze the main influencing factors of the total survival period(OS)and progression free survival(PFS)on mucoepidermoid carcinoma patients.Results(1)The positive expression rate of SEMA3 A,SEMA3B,p53 and Ki-67 in m ucoepidermoid carcinoma was 33.80%(24/71),28.17%(20/71),57.75%(41/71),43.66%(31/71)VS normal salivary glands 91.43%(32/35),85.71%(30/35),5.71%(2/35),(P<0.01).The differences were statistically significant(P<0.01).(2)In mucoepidermoid carcinom a,SEMA3 A expression was significantly correlated with age,lymph node metastasis,c linical stage;SEMA3B expression was significantly correlated with sex,age,tumor siz e,lymph node metastasis,clinical stage(P<0.05);p53 expression was significantly corr elated with tumor size,clinical stage(P<0.05);ki-67 expression was significantly correl ated with age,tumor size,lymph node metastasis,clinical stage(P<0.05);(3)There was a medium negative correlation between SEMA3 A and p53 expression(P<0.01,r=-0.474),there was a negative correlation between SEMA3 A and ki-67(r=-0.329,P=0.005),t here was a positive correlation between p53 and Ki-67(r=0.293,P=0.013).While,we did not find significant correlation among other proteins(P>0.05);(4)The results from Kaplan-Meier univariate survival analysis indicates that the overexpression of p53 may be a risk factor on overall survival of patients with mucoepidermoid carcinoma;the lo w expression of SEMA3 A and SEMA3 B,high expression of p53 and Ki-67,sex,lym ph node metastasis,tumor size,clinical stages are related factors affecting the mucoe pidermoid carcinoma patient's progression free survival(P<0.05).The results from Cox multivariate regression analysis showed that p53 may had no independent correlation withmay be a risk factor on overall survival(P>0.05);Ki-67 can predict progression fr ee survival as dependent factors in mucoepidermoid carcinoma patients(P<0.05).SEMA3 A,SEMA3B,p53 may had no independent correlation with progression free survival(P>0.05).Conclusions(1)The protein level expression of SEMA3 A,SEMA3B,p53 and Ki-67 are valuable in differentiating benign and malignant salivary gland tissues.SEMA3 A,SEMA3B are low expression and p53,Ki-67 are high expression.(2)The abnormal expression of protein SEMA3 A,SEMA3B,p53 and Ki-67 may provide reference for the diagnosis and prognosis of mucoepidermoid carcinoma of salivary gland.(3)Expression of Ki-67 protein may be an independent factor of progression free survival in patients with mucoepidermoid carcinoma of salivary gland.Overexpression of Ki-67 proteinmay be a risk factor.
