| Objective:To investigate whether IL-37 is expressed in vitreous and the expression in serum and vitreous of patients with proliferative diabetic retinopathy(PDR)by the way of measuring the content of IL-37 in the serum and vitreous body of patients who were diagnosed with proliferative diabetic retinopathy or non-diabetic ocular diseases.Methods:1.Subgroup : The subjects were divided into experimental group and control group.The subjects of experimental group were composed of 30 patients(30 eyes)with proliferative diabetic retinopathy who needed to be treated by vitrectomy.The subjects of the control group were made up of 30 patients(30 eyes)with non-diabetic who were diagnosed with idiopathic macular holes,macular premembrane or primary retinal detachment that required vitrectomy.2.Collection of serum samples:The fasting venous blood samples of the two groups were collected by the method of the median elbow vein and centrifuged at high speed.The serum samples were transferred into the Eppendorf tube and stored in the refrigerator at-80℃ to be tested.3.Collection of vitreous specimens:When the two groups of subjects underwent vitrectomy,before the opening of the perfusion tube switch,the 0.2ml of liquefied vitreous was extracted by 5ml syringe in the sterile state as the vitreous specimen.Then transferred into the Eppendorf tube and stored in the refrigerator at-80℃ for test.4.The Human IL-37 ELISA Kit(96 hole)was purchased from Wuhan Dr.de Company.Expression of all samples were assayed by enzyme-linked immuno sorbent assay according to the specification.5.Statistical analysis software SPSS 20.0 was used to analyze the results of general clinical data,biochemical indexes,blood routine indexes and IL-37 expression in the samples.Results:1.The general clinical data,biochemical indexes and blood routine indexes of the two groups were analyzed:There were no statistically significant differences in sex,age,low density lipoprotein,total cholesterol and lymphocyte count(P>0.05)between the two groups,but it has a statistically significant difference in fasting plasma glucose,high density lipoprotein,triglycerides,white blood cell count,neutrophil count and neutrophil lymphocyte ratio.2.The expression of IL-37 in serum and vitreous of patients in PDR group and control group could be detected.3.The expression of IL-37 in serum in PDR group was higher than that in control group,and the difference between them was statistically significant(P<0.05).4.The expression of IL-37 in the vitreous of the PDR group was higher than that in the control group.There was significant difference between the two groups(P<0.05).5.The expression of IL-37 in vitreous of the two groups was higher than that of IL-37 in blood.Conclusion:1.IL-37 is expressed in vitreous.2.IL-37 may be involved in the pathogenesis of diabetic retinopathy. |
