Objective:Asprosin is a new adipokine discovered recently,which traffics to the liver in vivo.Current studies have confirmed that it can stimulate the release of glucose in liver cells by activating the G protein-c AMP-PKA signaling pathway in the fasting state.At the same time,insulin resistance can increase the pathological level of Asprosin,while the specific antibody of Asprosin can improve the metabolic syndrome related hyperinsulinemia.Insulin resistance is one of the main pathogenesis of type 2 diabetes.However,it is not clear whether Asprosin is related to type 2 diabetes mellitus.The purpose of this study is to determine serum Asprosin levels in non obese and obese subjects with normal glucose tolerance and those with newly diagnosed type 2 diabetes,and to provide a new way of thinking for the treatment of obesity and type 2 diabetes.Methods:A total of 94 subjects(47 normal glucose tolerance population,47 T2DM)were enrolled in the study.According to the consensus(2016 Edition)of Chinese overweight/obese medical nutrition therapy expert(2016 Edition),the two groups were divided into non obese group(NOB)and obesity(OB)subgroup,respectively,with BMI?28kg/m~2 as the cutting point.Body and blood biochemical indexes of metabolism were tested,and circulating Asprosin was measured with ELISA kit,all data were processed by software SPSS20.0.The correlation between Asprosin level and other metabolic indicators was analyzed by linear correlation,multiple linear regression analysis,binary logistic analysis and so on.Results:(1)The level of Asprosin in group T2DM was significantly higher than that in group NGT[(387.974±126.417)vs(316.922±64.735)pg/ml,p=0.01].Asprosin level in T2DM-OB group was significantly higher than that in NGT-OB group[(381.069±105.490)vs(301.481±57.214)pg/ml,p=0.01].(2)Single factor correlation analysis showed that serum Asprosin level in T2DM patients was negatively correlated with HDL-C(r=-0.542,p<0.001),and positively correlated with FBG(r=0.786,p<0.001),HOMA-IR(r=0.467,p=0.009)and TG(r=0.706,p<0.001).After controlling BMI,WC and WHR,the level of serum Asprosin was positively correlated with FBG(r=0.485,p=0.002).(3)Taking serum Asprosin as dependent variable,FBG,HOMA-IR,TG and HDL-C as independent variables,multiple stepwise regression analysis showed that FBG(beta=0.384,p=0.009)and TG(beta=0.358,p=0.021)were independent risk factors of serum level.(4)To take obesity as the dependent variable,Binary Logistic regression analysis was used.There was no correlation between Asprosin and the occurrence of obesity.Binary logistic regression analysis was conducted on whether the occurrence of T2DM was the dependent variable,and Asprosin was correlated with the occurrence of T2DM,the OR was 1.039(95CI 1.007~1.072,P=0.018),after correcting the related factors,there was no significant correlation between Asprosin and the occurrence of T2DM.(5)By sex group,there was no statistically significant difference in serum Asprosin level in male group and female group,and Asprosin showed no significant gender in the NGT group and the T2DM group differences.Conclusion:The level of serum Asprosin in group T2DM was significantly higher than that in group NGT,and it was positively correlated with FBG,TG and HOMA-IR,and negatively correlated with HDL-C.FBG and TG were independent risk factors for serum Asprosin level.It is suggested that Asprosin as a new adipocytokine may participate in the pathogenesis of diabetes and play an important role in the pathophysiology of T2DM.
