Objective:Prepare the effecttive sites of Acanthopanax screen the best sites for the treatment of Parkinson's disease and ischemic stroke disease.Pro vide evidence for the pharmacological effects and clinical efficacy of Acanthopanax.In this way,we can expand to the application and development of Chinese medicinal herbs.Methods:Respectively put Acanthopanax roots and rhizomes of liquid water extractions in AB-8 macroporous resin,then use water and the volume fraction of 10%,30%,50%,70%,90%ethanol as the eluents.In addition to the total liquid extractions,7 kinds of effective sites were obtained;the Acanthopanax of leaves were extracted by 60%ethanol,put them in DM-130 macroporous resin.Washed with water and the volume fraction of 40%,70%,90%ethanol,coupled with the total extract,we acquire five kinds of pharmaceutically effective sites;Using superoxide(02-)anion clearance method,hydroxyl(OH-)radical scavenging method,DPPH method and remove ferrous reducing power experiments(FRAP)method comprehensive evaluation of their antioxidant capacity;Using PC-2 cells to establish MPP+ induced Parkinson's disease(PD)models,L-glutamate induced cell excitability toxicity damage model,hydrogen peroxide induced oxidative stress damage model,hypoxia hypoglycemia model.Give the effective sites of Acanthopanax,through the determined by MTT experiment for cell suivival.The effective site of the most protective ability of nerve cells was screened out under these damage models.lt will filter out parts of the pharmacodynamics experiments.The changes in behavioral activity were measured in C57/BL6 mice with spontaneous standing,climbing pole,roller,hanging in the experiment.Preliminary judge the screening site for the treatment of Parkinson's disease.Results:The washed with 30%ethanol of Acanthopanax roots and rhizomes and washed with 40%ethanol of Acanthopanax leaves have the strongest abilities to inhibit the damage from oxidation and and cell injury,protect the neurone from neurovirulence.the washed with 3 0%ethanol of Acanthopanax roots and rhizomes can protect neurone,we infer improved the Parkinson model of mices behavior abilities.Conclusions:Initially concluded that 30%of Acanthopanax roots and rhizomes and 40%of Acanthopanax leaves sites are the most active part of antioxidant activity in vitro sites,and have neuron protection,they can treat Parkinson's disease and ischemic stroke disease effectively.
