Screening For The Synergistic Substances In Sophora Alopecuroides L.combined With Lamivudine Against Hepatitis B Virus In Vitro

Objective:To explore the different extractions of Sophora alopecuroides L;the sophocarpine,the oxysophocarpine and Sophoridine;matrine,oxymatrine associated with lamivudine respectively in vitro inhibition of hepatitis B virus replication Comparative Study to examine aging,the amount of efficiency andstructure-activity relationship,and filter out the active ingredient for inhibiting viral Sophora alopecuroides.Methods:HepG2 2.2.15 cells in logarithmic growth phase selection of the tested drugs were bitter beans of various concentrations of ethanol extract;sophocarpine oxide,Sophora japonica fruitThe alkali sophoridine;matrine and lamivudine,oxymatrine and lamivudine;lamivudine as a positive control.For 3 days,6 days and 9 days later,the supernatant investigated the number of HBV DNA in HBsAg total extract,chloroform extract,butanol extract and the ethyl acetate and HBeAg and the cell supernatant.Results:1.Extracts from Sophora alopecuroides are the stronger the inhibition of antigen HBsAg,HBeAg and cell supernatant HBV DNA.2.Sophocarpine SRI alkali and oxysophocarpine strong inhibitory effect on HBsAg,HBeAg.3.Matrine in combination with lamivudine,oxymatrine in combination with lamivudine inhibition of HBsAg and HBeAg compared with both single-use about 10 times stronger.4.Matrine and oxymatrine comparison;sophocarpine contrast known oxysophocarpine,with the increase of polarity,drug inhibition of HBeAg.strong,with the decrease of polarity,drug of HBsAg inhibition.Conclusions:The findings by the subject preliminary judgment matrine,oxymatrine,the sophocarpine,the oxysophocarpine Sophoridine are anti-hepatitis B virus efficacy substances.The main component of matrine and oxymatrine in Sophora alopecuroides were together alone could significantly inhibit the antigen HBsAg and HBeAg compared with lamivudine with both.Provides a reference for clinical anti-hepatitis B virus and also laid the foundation for anti-hepatitis B virus drugs for the next development of bitter beans.
