| Platycodon grandiflorum, which can be used as a food, is the dry root of Platycodongrandiflorum(Jacq)A.DC which belongs to the Campanulaceae family. Modernpharmacology studies show that Platycodon grandiflorum has extensivepharmacolodical action and health protection function, which doomed that Platycodongrandiflorum has extensive development and application prospect. Although Platycodongrandiflorum has a wide range of application, most of the quality control methods areusing UV to determine the content of saponins which are the main chemical constituentsof Platycodon grandiflorum, and using HPLC to determine the content of platycodin-D.Platycodin-D as the single standard reference was used in the methods given above.Themethod determining the content of the single component can not control the quality ofthe medicine and preparation effectively. In order to improve the quality standard ofPlatycodon grandiflorum, this research carried out a series of studies on the basis ofpreliminary research.Objective: Based on the study of the chemical constituents of Platycodon grandiflorum,with an evaluation system of multiple index to establish a normalized quality standardsof Platycodon grandiflorum,to provide reference for the quality control of Platycodongrandiflorum and related products.Methods: Using physicochemical and TLC identification to conduct a comprehensivequalitative analysis of the chemical constituents in Platycodon grandiflorum.Using UVmethod to determine the contents of saponins, flavonids and sugar in Platycodongrandiflorum.Using HPLC method to determine the content of platycodin D anddeapio-platycodin D, luteolin,apigenin. Study on the HPLC fingerprint spectrum of20batches of Platycodon grandiflorum from different habitats. Results: The qualitve analysis results show that the chemical constituents ofPlatycodon grandiflorum contain polysaccharide,saponins,volatile oil and ones maycontain flavonoids,organic acid,protein,amino,exclude some chemical constituents ofthe flavonids,steroids,tannins,cardiac glycosides,alkaloids,anthraquinones,coumarin.TheTLC experiment confirmed that Platycodon grandiflorum contain saponins andflavonoids. The main chemical constituents of Platycodon grandiflorum was studied byquantitative analysis,UV method was established to determine content of totalsaponins,total flavonids and total polysaccharides.The scope of the total saponinscontent was2.34%3.27%, total flavonoids content was0.228%0.395%, content oftotal polysaccharide was18.2%46.3%.HPLC method was established to determine thecontents of saponins(platycodin D,deapio platycodin D) andflavonoids(luteolin,apigenin). The content ranges of platycodin D and deapio platycodinD were1.432.97mg·g-1and0.821.42mg·g-1.The luteolin content is in the range of154.18288.14μg·g-1,apigenin content was measured up to40.8μg·g-1,most did notdetected apigenin in the samples,respectively.On the basis,to establish a HPLC methodto study the fingerprint spectrum of19batches Platycodon grandiflorum fromShandong,Neimeng,Anhui.There are17common peaks in the standard HPLCfingerprint which can fully reflect consistency and overall characteristics of thechemical constituents in Platycodon grandiflorum.To establish a comprehensiveevaluation system of the quality of Platycodon grandiflorum.Conclusion: Establishment of UV method for the determination of total flavonids,totalsaponins,total polysaccharide and HPLC method for the determination of platycodinD, deapio platycodin D, luteolin, apigenin and fingerprint analysis are accurte andreliable,which can react the quality of Platycodon grandiflorum,completely. |
