Effect Of Locally Applying Iodine Tincture On Rabbits Free Flaps

BackgroundBreast cancer is the most common malignant tumors of women.It is also a common cause of cancer death in women.The comprehensive treatment based on surgery is still the basic treatment mode of breast cancer.Modified radical mastectomy is the most common surgical procedure used for breast cancer patients in China and other developing countries.Seroma formation is the most common postoperative complication of modified radical mastectomy.Up to now,there are no satisfactory measures to prevent and cure effusion.ObjectiveTo creat seroma formation models of NewZealand rabbit by applying normal saline and 2% iodine tincture on free flap and investigate the impact of locally applying 2% iodine tincture on healing wounding flap and reducing seroma formation.Methods32 New Zealand rabbits were randomly divided into four groups:A,B,C and D.The two groups of A and B were control groups,C and D groups were the treated groups.The flaps of the control groups were dealed with normal saline,while the treated groups were dealed with 2% iodine tincture.A and C groups were sacrificed on the third day after surgery,while B and D groups on the fifth day.We measured the results of weight,leukocyte count,neutrophil percentage,seroma volume of all groups and we tested the proliferation vessels and collagen fibers of the free flap tissue samples.All the data were analyzed with the corresponding statistical methods.Results1 Seroma volume was significantly decreased in C and D groups compared with A and B(P<0.05),in addition,seroma volume of C group was significantly decreased than that of B group(B vs C,P=0.005<0.05).2 The proliferation of collagen fibers and blood vessels of groups C and D were significantly higher than those of groups A and B(A vs C,P<0.001;B vs D,P<0.001),besides the proliferation of collagen fibers and blood vessels of groups C were significantly higher than that of B(B vs C,P<0.001).3 The proliferation of collagen fibers and blood vessels of group D was significantly higher than that of group C(P<0.001),while the proliferation of collagen fibers and blood vessels of groups B had no significant difference compared with that of A(P=0.520,P=0.208).ConclusionLocally applying 2% iodine tincture on free flap can reduce seroma formation,promote angiogenesis and collagen synthesis of free flap and accelerate wound healing process.
