Analysis On Efficacy Of Different Composed Of Kui Jie Shuang On Ulcerative Colitis

In this study,by orthogonal experimental design method,taking experimental UC rats as the research object to explore the dose-effect relationship and possible interactions of different components of the compound Kuijieshuang oral formula of Chinese medicine,determine the rational dosage of components and the best compatibility in the group prescriptions were used to screen out the best combination of traditional Chinese medicine for UC treatment.Clinical observations were performed to determine the clinical efficacy.The Kuijieshuang oral formula was divided into six factors,namely:Rougan Jianpi?Liver-Softening and Spleen-Nourishing?composition,Coptis chinensis,Radix militaris,Sophora flavescens,Phellodendron chinense,and honeysuckle.Firstly,each factor of the formula was taken at two levels,taking into account the interactions,and the L₈?2⁷?orthogonal table was selected for orthogonal test design and number 1 to 8 test drugs were obtained.72 healthy 8-week-old male Wistar rats were selected for experience.Except for the normal group rats,all the other groups were treated with dextran sulfate sodium?DSS?by intragastric administration to prepare the UC rat model.After the model was established,the prescription was given orally by referring to the orthogonal table.The course of treatment was 14 days.Observe the pathological scores of colon tissue in UC rats,and statistically analyze the test data,and finally select the best combination prescription for Chinese medicine treatment of UC,namely New Kuijieshuang oral prescription:Astragalus 30 g,Atractylodes 20 g,Poria 20 g,Ginger 12 g,raw licorice 12 g,eucalyptus 12 g,donkey-hide gelatin 12 g,angelica 20 g,white peony 30 g,ebony 12 g,corydalis 20 g,coke hawthorn 20 g,flavescens 10 g,medlar 30 g,berberine 6g,Treats 12 g,honeysuckle 15 g.Secondly,on the basis of experiment 1,50 wistar rats were selected.In addition to the 10 rats in normal group,40 rats that had been successfully modeled were randomly divided into 4 groups:Kuijieshuang Oral Formula Group,New Kuijieshuang Oral Formula Group and mesalazine group.A total of 4 groups,10 in each group,were treated with Kuijieshuang Oral Formula,New Kuijieshuang Oral Formula,and Mesalazineazine Enteric Tablets respectively.The course of treatment was14 days.After the course of treatment,the general conditions,DAI scores,serum CRP and IL-10 levels were observed in each group,and the colonic mucosal damage and histopathological scores were compared.The results showed that compared with the original prescription,New Kuijieshuang oral prescription can significantly reduce the CRP content and increase the IL-10 content in UC rats,and significantly improve the lesions of colon mucosa in UC rats,and the curative effect is significant?P<0.05?.Finally,we selected 60 UC patients in our hospital,dividing into two groups:treatment group?the New Kuijieshuang oral prescription,combined with traditional Chinese medicine enema?and control group?mesalazine enteric-coated tablets,combined with western medicine enema?,each group with 30 cases.After 12 weeks of treatment,we compared the clinical symptoms and endoscopic colon mucosal conditions.The results show that,the scores of diarrhea,sepsis,and abdominal pain in the two groups were significantly lower than before treatment,with statistically significant differences?P<0.05?;Chinese medicine group was better than Western medicine group in improving clinical symptoms such as diarrhea,pus and blood stasis,and the difference was significant?P<0.05?);There was no significant difference in the symptoms of abdominal pain between the Chinese medicine group and the Western medicine group.After treatment,both groups could significantly improve the condition of colonic mucosa,and the Chinese medicine group was better than the western medicine group,but the difference was not statistically significant?P>0.05?.Conclusion:New Kuijieshuang Oral Prescription can significantly reduce the content of CRP in UC rats,increase the content of IL-10,and improve the condition of colonic mucosain.And,New Kuijieshuang Oral Prescription can significantly improve the clinical symptoms of UC patients,improve the mucosal lesions of the colon.
