With the development of third generation iron supplement,researchers began to extract polysaccharides from plants and complexed with iron to develop polysaccharide iron complex.However,there are still problems such as low iron content and low solubility.For this purpose,the polysaccharides extracted from the Thesium,which complexed with iron after removing impuritiesare was used to prepare the third generation of iron supplement.The iron content,process optimization,structure,physicochemical properties and dissolution characteristics were studied.1.By using alcohol precipitation technology to extract polysaccharides from Thesium,the content of Thesium polysaccharides was 52.82% after decolorization,deproteinization and dialysis.The Thesium polysaccharides iron complex(TPIC)was prepared by complexed polysaccharides which extracted from Thesium with iron.Single factor experiments showed that citric acid,iron trichloride,sodium hydroxide and sodium carbonate had significant effects on the synthesis of polysaccharides iron.The orthogonal experiment results showed that the best synthesis condition as follow: 0.15 mol/L sodium citrate 14 mL,5% sodium carbonate 4 mL,0.45 mol/L ferric chloride 18 mL,with 10%sodium hydroxide to adjust pH8.The iron content is 52.86% under this condition.The influence order as follow: iron trichloride addition>pH>sodium citrate addition>sodium carbonate addition.The results of the SPSS significance analysis showed that the sodium citrate addition(A)had a significant effect on the yield(0.042 < 0.05),and the pH value(C),which is the sodium hydroxide addition had a significant effect on the iron content(0.038 < 0.05).2.The structure of the Thesium polysaccharides iron complex was analyzed.Through400~700nm UV Vis Spectrum Analysis,it was concluded that the Thesium polysaccharides was complexed with ferric iron.Through the infrared spectrum analysis,the result suggested that Thesium polysaccharides iron complex was composed of Thesium polysaccharides and iron.There exists hydrogen bonding interaction between the polymeric iron core and the polysaccharide.The hydroxyl-OH in the polysaccharides was involved in the complexation.The carbonyl C=O bond of the carboxylic acid in the polysaccharides may be involved in the complexation of the polysaccharides and the trivalent iron.The X-ray diffraction analysis showed that the X diffraction pattern of the Thesium polysaccharides iron complex is characterized by a weak broad diffraction peak atthe space between d=0.25 nm and 0.15 nm.It is preliminarily determined that the composition of the iron polycore of the polysaccharide iron is a unit lattice contraction?-FeOOH structure.3.The physicochemical properties of the Thesium polysaccharides iron complex were analyzed.The powder of the Thesium polysaccharides iron complex was red brown,no special odor,good water solubility,insoluble in anhydrous ethanol and other organic reagents.There was no free ferric ion in the Thesium polysaccharide iron complex.The Thesium polysaccharide iron complex was stable both in air and water.When the pH of Thesium polysaccharides iron complex solution was changed from 3 to 12,no precipitation was formed,indicated that it was soluble and stable under physiological conditions.4.The iron content of the Niferex polysaccharide iron capsule was 45.86%,which was basically consistent with the labeled content of 46%.It was proved that o-phenanthroline spectrophotography used in this experiment was viable.The dissolution rate of the Thesium polysaccharides iron complex and Niferex polysaccharide iron capsule was larger in 1h,and the dissolution rate in 2-5h continued increasing.The dissolution after 5h was basically stable and dissolved completely in 6h.By studying the effect of pH and Vc on the iron dissolution rate of the Thesium polysaccharides iron complex and the Niferex polysaccharide iron capsule,the results showed that the dissolution rate of pH2 was higher than that of pH6.8.The content of 10% and 20%Vc had less effect on iron dissolution under the same pH value.In a word,the iron dissolution rate of the Thesium polysaccharides iron complex was higher than Niferex polysaccharide iron capsule at the same time.
