Objectives Through the comparison of different doses of buckwheat flavonoids on diabetic nephropathy rats after fasting blood glucose,blood lipid,serum creatinine,urea nitrogen,24 hours urine protein and renal tissue MDA and SOD content,to investigate the protective effect of buckwheat flavonoids on diabetic nephropathy rats and its mechanism,and to find the appropriate dose of buckwheat flavonoids on diabetic nephropathy rats.Methods 90 healthy SD rats were selected,5 females and 5 male rats were randomly selected as normal control group.The rest 80 were given four oxouracil 150 mg/kg/ times,one week and three times for diabetic nephroppathy rats.The model rats were randomly divided into model control group,irbesartan group(17mg/kg)and buckwheat yellow Ketone in low,medium and high dose group of 50,100,200 mg/kg.The buckwheat flower leaves were completely crushed and buckwheat flavonoids were extracted.Different doses of buckwheat flavonoids and erbesartan groups were given corresponding drugs,and normal saline and model control group were given the same volume of normal saline(NS).Each group was administrated 1 times a day for 12 W continuously.The FBG and 24 hour urine albumin levels were measured every 3 W.The serum urea nitrogen(BUN),serum creatinin (SC_r),total cholesterol(TC),triglyceride(TG)and FTS,AGEs,SOD and MDA in the kidney tissues were determined after the last dose of 2H.Meanwhile,2 rats in each group were subjected to light microscopic analysis.Results Compared with normal control rats,FBG in model control group and FB group were significantly higher than those in normal control group(16.7mmol/L).Compared with the model control group,the FBG,24 h urine protein,SCr,BUN,TG,TC,FTS,AGEs and MDA decreased in different degrees after continuous administration of FB,and decreased to the lowest level at the 12 week after the administration,and the SOD increased in different degrees,but the high dose group did not obtain lower FBG,24 hour urine protein,SCr,BUN,TG,TC et ca than middle dose group,but also did not obtain higher SOD.Conclusions Different doses of buckwheat flavonoids can protect diabetic nephropathy rats by decreasing blood sugar,reducing blood lipids,improving renal function and 24 h protein,and its protective effect is related to the antioxidant effect.The appropriate dose of buckwheat flavone for diabetic nephropathy rats is 100mg/kg.
