Objective Titanium and titanium alloy have so many excellent characteristics such as good biocompatibility,high hardness and anti-corrosion.They are widely used for implanting in clinic,but the ability of combining with bone is weak.We often modify their surfaces with physical,chemical and biological methods to develop their ability of combining with bone.This experiment is about the effect of micro-arc oxidation of pure titanium and calcium-loaded coating on cell biological activity by alkali steaming,ultraviolet sterilization,high temperature and pressure sterilization.At last searching for the best modified method and the best method of sterilization among these kinds of modified coatings.Methods This experiments are totally divided into four groups.Micro-arc oxidation of pure titanium(Group A)in the solution which is made up of trisodium phosphate and sodium silicate mostly.Micro-arc oxidation of pure titanium and calcium-loaded coating(Group B).Micro-arc oxidation of pure titanium and calcium-loaded coating on cell biological activity by alkali steaming and ultraviolet sterilization(Group C)and high temperature and pressure sterilization(Group D).Macro observation and SEM observation show the morphology and colour of surface.Making use of CCK-8 reagent to test Adhesion and proliferation of mouse osteoblasts among above four groups.Making use of ALP and BCA reagent to test the active of mouse osteoblasts.Making use of confocal laser scanning microscope to observe the spread area of cytoskeleton and the number of microfilaments and pseudopodium of mouse osteoblasts.Results Macro observation of micro-arc oxidation of pure titanium shows dark yellow visibly and uniform surface.Macro observation of micro-arc oxidation of pure titanium and calcium-loaded coating shows dark blue visibly and uniform surface slightly.Macro observation of micro-arc oxidation of pure titanium and calcium-loaded coating on cell biological activity by alkali steaming and ultraviolet sterilization shows dark blue grey visibly and uneven surface slightly.Macro observation of micro-arc oxidation of pure titanium and calcium-loaded coating and high temperature and pressure sterilization shows dark grey visibly and uneven surface and coloboma slighty.SEM observation of the surface of micro-arc oxidation of pure titanium is porous.SEM observation of the surface of micro-arc oxidation of pure titanium and calcium-loaded coating exits granular calcium particle after deposited calcium.SEM observation of the surface of micro-arc oxidation of pure titanium and calciumloaded coating on cell biological activity by alkali steaming and ultraviolet sterilization takes shape average micro-nano particles.SEM observation of micro-nano morphology of micro-arc oxidation of pure titanium and calcium-loaded coating and high temperature and pressure sterilization is destroyed.The experiments of cell adhesion and proliferation shows micro-arc oxidation of pure titanium and calcium-loaded coating on cell biological activity by alkali steaming and ultraviolet sterilization shows the number of Absorbance(optical density)are lager than the other three groups.The experiment of alkaline phosphatase to cell activity shows cell activity of micro-arc oxidation of pure titanium and calcium-loaded coating on cell biological activity by alkali steaming and ultraviolet sterilization is the largest.Cytoskeleton observation shows the spread area of cell and the number of microfilaments and pseudopodium of micro-arc oxidation of pure titanium and calcium-loaded coating on cell biological activity by alkali steaming and ultraviolet sterilization are the largest and intercellular connection is more closer.Conclusions the cellular compatibility of micro-arc oxidation of pure titanium and calcium-loaded coating on cell biological activity by alkali steaming and ultraviolet sterilization is best among four groups.
