Objective: The terahertz spectroscopy(THz)technology has been widely used in communications,aerospace,medical,and medical equipment for its advantages of simple pretreatment process,low resource consumption,environmental protection,short detection cycle,simple detection process,and flexible data processing algorithms.Drug testing and other areas,its application in the study of traditional Chinese medicine is still at its infancy.This thesis aims to explore the applicability of terahertz time-domain spectroscopy in the identification of three medicinal materials of Angelicae dahuricae Radix,Chuanxiong Rhizoma,Notopterygii Rhizoma et Radix,in order to establish a new method for the identification of traditional Chinese medicine THz,and to supplement Chinese medicine identification methods.Methods: In this study,terahertz time-domain spectroscopy was introduced into the identification of authentic Chinese herbal medicines.The study included the application of this technology to the identification of Angelicae dahuricae Radix,Chuanxiong Rhizoma,Notopterygii Rhizoma et Radix.The traditional Chinese medicine identification method is used to ensure the differences in the samples.The terahertz time-domain spectroscopy technique is used to identify the different harvested and processed medicines of different traditional Chinese medicines and compare the respective advantages of the two methods in order to find one.A impler and more efficient technique has been applied to the identification of Chinese herbal medicines to make up for the deficiencies of existing methods.Results: The results of this study are as follows:(1)The application of terahertz time-domain spectroscopy in the identification of Angelicae dahuricae Radix unsmoked and smokedThe traditional Chinese medicine identification method was used to identify the characteristics of samples including Angelicae dahuricae Radix unsmoked and smoked.The content of leachables and the index components(white angelica,imperatorin and isoimperatorin)were determined.Content determination and cluster analysis and principal component analysis based on HPLC fingerprinting showed thatthere were significant differences in traits,leachate content,and HPLC fingerprinting between Angelicae dahuricae Radix unsmoked and smoked and most of the samples were able to follow Smoked or not individually clustered.Terahertz time-domain spectroscopy was used to extract the terahertz spectral information of 19 batches of Angelicae dahuricae Radix.The PCA algorithm was used to process the data.The results showed that the samples of smoked ones and unsmokeed ones were gathered on the three-dimensional principal component projection map.There are two types and no intersection,that is,the recognition rate of unsmoked ones and smoked ones is100%.Comparing the results of the two methods,it shows that terahertz time-domain spectroscopy can not only extract the information of the dissimilar chemical constituents of unsmoked S.fruticosa and S.fruticosa,but also has higher extraction efficiency.This method is able to solve the problem of identification of Angelicae dahuricae Radix unsmoked and smoked.(2)Terahertz time-domain spectroscopy applied to the identification of different specifications Chuanxiong Rhizoma harvested in different timeThe traditional Chinese medicine identification method was used to identify the traits,extracts,and ferulic acid content in Chuanxiong Rhizoma harvested in different time,as well as the cluster analysis and principal component analysis based on HPLC fingerprints.There were significant differences in the traits and HPLC fingerprinting of milk thistle.The cluster analysis showed that most of the Chuanxiong clams clustered into one group and most of the Chuanxiong Rhizoma harvested in different time clustered into one group.Using the terahertz time-domain spectroscopy technique to detect these samples,we found that the projection method was used to extract the spectrum and extract the principal components to reduce the dimension.In the low-dimensional data visualization,the three-dimensional view of the principal components showed Kagawa samples.In the image,there are both separate and intersecting parts in the sample.Finally,the classifier is trained to build a model by selecting an appropriate classifier and method.The classification results are used to distinguish most of the Chuanxiong Rhizoma harvested in different time,nearly 100%.So the result shows that the technique is able to distinguish the Chuanxiong Rhizoma harvested in different time.(3)Terahertz time-domain spectroscopy applied to the identification of Notopterygii Rhizoma et Radix with different specifications and grades of Notopterygii Rhizoma et RadixIn the earlier period,the research group systematically studied Notopterygii Rhizoma et Radix with different specifications and grades of Notopterygii Rhizoma et Radix using traditional Chinese medicine identification methods.The content of leachables,volatile oil content,composition and relative content of Notopterygii Rhizoma et Radix with different specifications and grades of Notopterygii Rhizoma et Radix,total content of locust alcohol and isoimperatorin were determined.Anti-inflammatory effects,analgesic effects and other aspects were examined.The results of the study showed that there are differences in the livelihood of Notopterygii Rhizoma et Radix with different specifications and grades of Notopterygii Rhizoma et Radix,among which silkworm cocoons are significantly superior to other specifications of cockroaches;HPLC fingerprints are available for different product specifications.In the study of the map,it was found that the HPLC of the cultivated alfalfa had obvious differences and could be identified.However,HPLC chromatograms of other alfalfa varieties were very similar and could not be identified.The terahertz time-domain spectroscopy technique was applied to the identification of different commodity specifications for deficiencies.Under this technique,different live product samples of different product specifications can be clustered relatively more regularly according to commodity specifications.However,there are still a large number of different types of products.In the intersecting part,only the clustering of the silkworm cocoon is relatively clear,that is,the terahertz time-domain spectroscopy technology can realize the complete recognition of the cocoon and the fuzzy identification of other product specifications.Although this method also does not realize the identification of all the different commodity specifications in the market,it to some extent makes up for the lack of chemical methods and provides new ideas and methods for the identification of different commodity specifications.Conclusion: The terahertz time-domain spectroscopy technology can,to a certain extent,realize the identification of Angelicae dahuricae Radix,Chuanxiong Rhizoma,Notopterygii Rhizoma et Radix.Although the established model needs to be further tested and optimized,its feasibility and applicability in the identification of these three Chinese herbal medicines are undoubted.This method is also expected to be used as a new,highly efficient and environmentally friendly method for the identification of other Chinese medicines.In the middle,enrich the methods and ideas of traditional Chinese medicine research.
