Background:Oral Lichen Planus,OLP is a T-cell-mediated chronic inflammatory disease with unknown etiology.The cause of oral lichen planus remains unknown.It is reported that the oral lichen planus becomes malignant rate is 0.4%-2%[1].The occurence and development of the oral squamous cell carcinoma may come from a sample of the tumor micro-environment,including anaerobic,inflammation,immune and acidic environments,a great deal of promotion of oral lichen planeus for cancer patients.The research of the staus of oral cancer has a very important significance for the prevention of the occurence and development of oral cancer.Oral squamous cell carcinoma,OSCC is the most common malignant tumor in oral and maxillofacial cavity.Oral squamous cell carcinoma is mostly developed by precancerous lesions,such as white spots and lichen moss[2].most patients diagnosed the oral squamous cell carcinoma the time already was?Time or?Time[3],for the life of patients with a serious threat,the average five-year survival rate is only 53%[4].So we can prevent the cancelation process if we pre-sentence the occurrence of the oral squamous cell carcinoma.The CD133 protein is one kind of cross membrane protein which discovered on the human hematopoietic stem cell.It was original considered as a blood stem cell specificity.Some studies have shown that CD133 may participate in the occurrence of oral cancer,abnormal CD133 expression is associated with malignant change of oral leukoplakia[5].Very few articles on the relationship between CD133 and carcinogenesis of oral mucosaObjective: To study the changes of protein expression CD133 in three samples of normal oral mucosa,oral lichen planus tissue and oral squamous cell carcinoma,and evaluate the correlation between the expression of CD133 and the risk of oral lichen planus transform into the oral squamous cell carcinoma,for later provide further evidence for research and treatment of cancer.Materials and Methods:Collects 10 cases of normal oral mucosal tissue specimens;45 sample wax block of the oral lichen planus organization;21 cases of tissue wax block of oral squamous cell carcinoma(Oral mucosal squamous cell carcinoma tissue wax blocks were divided into three groups according to pathological grade: high-differentiation group,medium-differentiation group and poorly differentiated group.),there are 5 cases of oral lichen moss tissue and 5 cases of oral squamous cell carcinoma were tissue specimens of the same patient at different times,and the 5 patients were diagnosed with oral lichen planus cancer as oral squamous cell carcinoma by clinical and pathological science.Adopt the method of immune histochemical dyeing on the tissue pieces.Immune histochemical results using the semi-quantitative method for interpretation,record the staining intensity and dyeing cells proportion,two multiplied to obtain final score,score according to distinguish between the following four levels: 0 can be divided into negative,1-3 into the weak positive,4-8 into moderate positive,9 or higher is divided into strong positive[6].The obtained data carries on statistics analysis with SPSS21.0,comparisons between different groups using level data rank and inspection,three groups of tests with ?2.The examination standard alpha?takes 0.05,P<0.05 as there are statistically significant differences.Results:1.CD133 expression in oral normal mucosa tissues:In the normal mucous membrane expresses rate is 0,10 example normal mucous membranes has not seen CD133 after the dyeing the expression.2.CD133 in oral lichen planus expressed in the organization:45 cases of oral lichen planus organization,26 cases of negative,13 cases characterized by weak positive,6 cases of moderate positive,positive rate was42.0%.Oral squamous cell group positive expression is higher than oral normal mucosa tissues.According to the statistical analysis of the experimental group compared with normal oral mucous membrane group,the difference has statistically significance(P<0.05).3.The expression of CD133 in oral squamous cell carcinoma tissue:In oral squamous cell carcinoma,CD133 central assumes the strong masculine expression in the cancer nest.In the 21 cases of oral squamous cell carcinoma tissues,7is in the negative,6 is shown to be a weak positive,6 showed a moderate positive,2 to demonstrate a strong positive,positive rate of 66.7 percent.Along with the normal mucous membranes of the mouth and oral lichen planus,this group shows statistically significance equally.(P<0.05).4.Expression of CD133 in the tissues of 5 patients treated with oral lichen planus as oral squamous cell carcinoma:One patient's oral lichen planus organizations appear to be negative,oral scale-cell cancer organizations in the form of moderate positive;2 patients for oral lichen planus organizational performance to be a weak positive,oral scale-cell cancer organizations in the form of moderate positive;2 patients for oral lichen planus organizational performance to be a weak positive,oral squamous cell carcinoma organizations in the form of a strong positive.In the pathological change process of the patient,the content of CD133 is rising.5.Oral mucosal squamous cell carcinoma tissues were divided into three groups according to pathological grade: high-differentiation group,medium-differentiation group,and poorly differentiated group.A ?2 test was performed between the three groups.The ?2 value was 14.193 and the P value was 0.028(<0.05).The difference between the three groups was statistically significant.Conclusion:1.In normal oral mucosa tissues,oral lichen planus tissue and the oral squamous cell carcinoma,the positive expression rate sees an increasing trend.2.Expression of CD133 in the tissues of 5 patients treated with oral lichen planus as oral squamous cell carcinoma:In the pathological change process of the patient,the content of CD133 is rising.3.The quantity of protein expression of CD133 can be used as evaluation index of oral squamous cell carcinoma It can be inferred that CD133 plays a role in the changes of normal oral mucosa ? oral lichen planus ? oral squamous cell carcinoma.4.The expression level of CD133 is related to the differentiation of oral squamous cell carcinoma.The higher the pathological differentiation of oral squamous cell carcinoma,the lower the positive expression of CD133.5.CD133 protein is associated with the carcinogenesis of oral mucosa lichen planus.For oral mucosal lichen planus with positive expression of CD133,clinical interventions should be increased.For example,oral mucosa lichen planus can be actively treated to prevent oral mucosa lichen planus cancer.
