bjective: Deep venous thrombosis of lower limbs is a common venous vascular lesion that can be prevented difficultly in the clinical.Chinese medicine has definite effects in preventing deep venous thrombosis of lower limbs.But there is no effective,convenient and useful Chinese medicine compound preparation.In this paper,the experiment of thrombus in rats was designed by orthogonal design table.Select relevant effective indicators for testing,so as to screen out the optimal formula composition of the Fang Shuan Fang to prevent Deep venous thrombosis.Methods: According to the orthogonal design table of L12(211),Fang Shuang fang was divided into 12 groups(the Chinese medicine prescription splitting groups numbered 1-12).Meanwhile,blank control group,model group,sham operation group and warfarin sodium group were designed.160 SD rats were selected,half male and half female.They were adaptive feed for 7 days after repurchase.Female and male rats were separately numbered and randomly divided into 16 groups according to the random number table,with 10 rats in each group and 5 males and 5 females.Seven days before the modeling,the rats in each of the Fang shuang fang splitting group were given the corresponding dose of the Chinese medicine decoction.The rats in the blank control group,model group and sham operation group were given normal saline.And the rats in the warfarin sodium group were given warfarin sodium solution by gavage(0.2mg/Kg).The rats in each group were given the corresponding drug once a day by gavage for 7 days.We begin to build the model of Vein thrombosis in rats after the last feeding thirty minutes,remove the Vein thrombosis after modeling 6 hours.Then we weigh the dry and wet weights of Vein thrombosis.About 2.7ml of blood was drawn from the abdominal aorta of the rats before removing the Inferior Vein thrombosis,which was stored in a coagulation function test tube.Then add 3.8% Sodium citrate into the blood.The blood was centrifuged at a speed of 3000r/min for 15 min.Separat the plasma from the blood,then determine APTT,PT,TT and FIB according to the Operating instructions.All data were analyzed using SPSS 17.0 software.(3)Results:(1)There was no DVT in the blank group and the sham group,while all rats in the model group developed thrombosis,and the thrombosis rate was 100%.Compared with the model group,the wet mass and dry mass of thrombus of the The original prescription of Fang Shuan fang both decreased(P<0.05).(2)Compared with the blank group,PT,APTT and TT of the model group showed a significant shortening trend(P<0.05),FIB showed a increasing trend(P<0.05).Compared with the model group,PT,APTT and TT of rats in the original Fang shuan fang group were longer(P<0.05),and FIB was lower(P<0.05).(3)Factor A(astragalus)had significant effects on prolonging PT and APTT of rats and reducing wet weight and dry weight of thrombus(P<0.05).Factor B(dang shen)had statistical significance in reducing wet weight and dry weight of thrombus(P<0.05).Factor C(salvia miltiorrhiza)had significant effects on prolonging PT,APTT,TT and reducing wet weight and dry weight of thrombus in rats(P<0.05).Factor D(radix paeoniae alba)had significant effects on prolonging APTT and reducing wet weight and dry weight of thrombus in rats(P<0.05).Factor F(peach kernel)had significant effects on prolonging PT,APTT and TT and reducing FIB,wet weight and dry weight of thrombus in rats(P<0.05).Factor G(safflower)played a significant role in prolonging PT and APTT in rats and reducing FIB,wet weight and dry weight of thrombus in rats(P<0.05).Factor H(chuanxiong)had significant effects on prolonging APTT and TT of rats and reducing wet weight and dry weight of thrombus(P<0.05).Factor I(Zhi shi)had a negative effect on reducing FIB(P<0.05).Factor J(rhubarb)had a negative effect on prolonging PT in rats(P<0.05).Factor K(Niu xi)played a significant role in prolonging PT and TT and reducing wet and dry weight of thrombus in rats(P<0.05).Conclusion:(1)Deep venous thrombosis(DVT)animal models can be successfully established by ligation of inferior vena cava.(2)The original prescription of Fang Shuan fang can effectively prevent DVT,and its prevention mechanism may be to affect the endogenous and exogenous coagulation system activity.(3)The Concise formula of Fang shuan fang is constituted by astragalus,salvia miltiorrhiza,radix paeoniae alba,angelica sinensis,ligusticum,chuan xiong,peach kernel,safflower,and niu xi.
