ObjectiveThrough clinical trial and experimental reserch to study the effect of Shenqi Guben Fang ultrafine granular powder on bone marrow after chemical treatment.Methods1.Animal Experiment80 KM male mice,component to 5 groups randomly:Blank Group,Model Group,Shenqi Guben Fang Equal to Clinical dosage Group,Half to Clinical dosage Group,One-fourth to Clinical dosage Group.Make up one group with 16 mice and mark with picronitric acid.Except for the Blank Group,the rest of the four groups were injected the Cytoxan(100mg/kg·d)into the abdomen for 3 days.The Blank Group were injected the same dose of normal saline into the abdomen.The Blank Group and the Model Group were administrated normal saline by means of intragastric administration when the other three groups with the ultrafine granular powder in different dose with lml/lOg weight feeding into stomach for 7days.Observe index sign:weight,routine blood test,lympho T subset cell.2.Cell ExperimentsPrepared different concentrations of Shenqi Guben Fang ultrafine granular powder(0.1mg/ml?0.2mg/ml?0.3mg/ml?0.4mg/ml?0.5mg/ml),and used it to treat on the cell model of A549.MTT method were used to assay the A549 activity.Observe index sign:Optica.l density,0D.3.Clinical trialsCollected 40 patients who has been diagnosised with non-small cell lung cancer.They would use the chemotherapy regimens like DP(Docetaxel+Cisplatin)and AP(Pemetrexed+Cisplatin).At the same time,these 40 patients were divided into two groups randomly.One was to be treated with the Shenqi Guben Fang ultrafine granular powder(30g,blunt,bid)for 56 days.The other one was to be treated with the Chinese Herbal Pieces of Shenqi Guben Fang(120g,oral,bid)for the same days.Input the data and compare these indicators,including the physical condition and the symptoms,the occurrence of bone marrow inhibition before and after chemotherapy,the number of immune parameters including CD4+,CD8+and CD4+/CD8+.Results1.Animal ExperimentWeight:Contrasted with the Model Group,the Shenqi Guben Fang ultrafine granular powder in different dose could help to put on weight(P<0.05).Routine blood test:Contrasted with the Model Group,the Shenqi Guben Fang ultrafine granular powder in different dose could increase WBC,PLT(P<0.05),and there is no statistical difference among these three groups in WBC,PLT.The group which is equal to the clinical dose could increase HGB(P<0.05).Lympho T subset cell:Contrasted with the Model Group,the Shenqi Guben Fang ultrafine granular powder that equivalent to clinical dose could increased CD4+and CD4+/CD8+,decreased CD8+(P<0.05).The 1/2 and 1/4 clinical dose group couled decreased CD8+,but there were not statistically significant in increasing CD4+ and CD4+/CD8+(P>0.05).2.Cell ExperimentsCompared with the Control Group,each groups restrained the A549' s growth(P<0.05),except for the 0.1mg/ml and 0.2mg/ml of the 24h Group(P>0.05).Compared with the 24h Group,the OD of each 48h and 72h groups were remarkably down(P<0.05).3.Clinical trialsPeripheral blood:WBC is higher than before treatment(P<0.05),but there was no statistically significant difference in PLT and HGB before and after treatment(P>0.05).Between 2 groups,WBC,PLT and HGB didn't have statistically significant difference.In the T-lympocyte subsets,CD4+ is higher than before treatment(P<0.05);but there was no statistically significant difference in CD8+and CD4+/CD8+before and after treatment(P>0.05).Between 2 groups,CD4+,CD8+ and CD4+/CD8+didn' t have statistically significant difference.In terms of the marks of symptoms,the marks of fatigued spirit and weak,and loss of appeitite were decreased with the statistically significant(P<0.05).The marks of soreness and weakness of waist knees,and insomnia and dreamful sleep were without the statistically significant(P>0.05).There were no statistically significant in the marks of the symptoms between these two groups(P>0.05).Conclus ion1.Animal ExperimentThe three kinds of dose of Shenqi Guben Fang ultrafine granular powder not only conuld increased peripheral blood cells of WBC,PLT and the weight but also could decrease CD8+ of the bone marrow model mice.The Shenqi Guben Fang ultrafine granular powder that equivalent to clinical dose not only could increase HGB,but also could increase CD4+and CD4+/CD8+which could help to improve immune function.2.Cell ExperimentsShenqi Guben Fang ultrafine granular powder can inhibit the growth of A549 cells,which has a concentration and time dependency.3.Clinical trialsShenqi Guben Fang ultrafine granular powder and the Chinese Herbal Pieces one could improve the symptoms like fatigued spirit and weak,and loss of appeitite,but no effect on the symptoms of soreness and weakness of waist knees and insomnia and dreamful sleep.In this part,The 40 patients have the same respond to these two forms of drug treatment.Shenqi Guben Fang ultrafine granular powder and the Chinese Herbal Pieces one could increase the number of WBC to improve the situation of bone marrow.And the 40 patients have the same respond to these two forms of drug treatment.Shenqi Guben Fang ultrafine granular powder and the Chinese Herbal Pieces one could increase the percentage of CD4+,help boost immunity.In this part,the 40 patients have the same respond to these two forms of drug treatment.Under the premise that the dosage of the ultrafine granular powder is only half of the Chinese Herbal pieces,the two dosage forms have the same effect,which indicates that the dose effect relationship of ultrafine granular powder is better than that of the Chinese Herbal pieces.
