ObjectiveTo validate the effectiveness of continuous thermal dressing in wound healing and to explore the effects of continuous thermal dressing in wound repairing and related mechanisms.MethodsForty C57BL/6 mice were divided into two groups according to random number table with 20 mice in each group.Full-thickness skin defect models with a diameter of 10mm were constructed and a silicone ring sutured on to prevent contracture of wound on both groups.For the experiment group,constant heat was maintained at 36? and the temperature data was recorded continuously.For the control group,the thermal dressing was turned off and the temperature data of the wound was recorded continuously.On the 7th,10th and 14th day post-surgery,4 mice were randomly taken from each group and the wound healing rate was calculated after an overall photo of the wound was taken;the wound tissue was taken,and the histopathological changes were observed through hematoxylin-eosin staining;the expression and distribution of CD31 and a-SMA were observed through immunohistochemical staining;real-time florescence-based quantitative reverse transcription-polymerase chain reaction was used to detect the mRNA expression of CD31,a-SMA,VEGF,collagen I,collagen ?,TL-6 and IL-10 in wound tissues;the protein expression of CD31,a-SMA,VEGF,collagen ?,collagen ?,IL-6 and IL-10 were detected through Western blotting.After normality test and homogeneity of variance analysis was done on the data,Student's t-test was used to compare the two groups at each time point.Results(1)The overall results showed that the wound area of the experiment group was smaller than that of the control group on the 10th and 14th day post-surgery,with the statistical difference of p<0.05.On the 7th,10th and 14th day post-surgery,the wound healing rate of experiment group was significantly higher than that of the control group,with statistical significance of p<0.05.(2)The results of hematoxylin and eosin(HE)staining showed that the regenerated epithelial and granulation tissues in the experiment group were thicker than those in the control group on the 10th and 14th day post-surgery.Fibroblasts,neovascularization and collagen deposition in the experiment group was significantly higher than that in the control group.(3)Comparing the results of CD31 and ?-SMA immunohistochemical staining and blood vessel count,the number of neovascularization and mature blood vessels in the experiment group on the 10th and 14th day post-surgery was higher than that of the control group(p<0.05 or P<0.01).The molecular expression of mRNA's CD31,a-SMA and VEGF in the wounds of both groups increased gradually after surgery.On the 7th day post-surgery,the mRNA expression of a-SMA and VEGF in the wounds of the experiment group was higher than that of the control group(p<0.05);on the 10th day post-surgery,the mRNA expression of CD31,a-SMA and VEGF in the wounds of both groups was higher than that of the control group(p<0.05).On the 14th day post-surgery,the expression of CD31 and a-SMA in the experiment group was higher than the control group(p<0.05 or P<0.01).The results of protein electrophoresis were consistent with qRT-PCR and immunohistochemical results.(4)The mRNA expression of collagen I and collagen ? in wounds of both groups gradually increased with time after surgery.On the 7th and 14th day post-surgery,the mRNA expression of collagen I and collagen ? in the experiment group was higher than the control group(p<0.05 or P<0.01);on the 10th day post-surgery,the mRNA expression of collagen I in the experiment group was higher than the control group(p<0.01).The protein expression levels of collagen I and collagen ? in both groups on the 7th,10th and 14th day post-surgery increased gradually with time.The protein expression levels of the experiment group were higher than the control group at each time point.(5)On the 7th,10th and 14th day after operation,the expression of IL-6 mRNA in the wound tissues of both groups decreased gradually post-surgery.On the 7th day post-surgery,the expression of IL-6 mRNA in the experiment group was less than that in the control group(p<0.01);on the 14th day post-surgery,the expression of IL-6 mRNA in the experiment group was significantly lower than that in the control group(p<0.05).On the 7th,10th and 14th day post-surgery,the expression of IL-10 mRNA in both groups decreased gradually with the expression in the experiment group higher than the control group(p<0.05).Western blotting was used to detect the protein expression changes of IL-6 and IL-10 on the 7th 10th and 14th day post-surgery,which was consistent with the results of qRT-PCR.ConclusionContinuous thermal dressing on the wound can promote the healing of full-thickness skin defect wounds in mice.Its mechanism may be related to promoting skin vascularization,epithelialization,collagen deposition and inhibiting inflammation in the mice.
