Preparation Of Chitosan-collagen Sponge And Its Effect On Wound Healing

ObjectiveWe prepared a chitosan-collagen sponge and studied its cytotoxicity,antibacterial property,moisture retention and its effect on wound healing.MethodsChitosan-collagen sponge was freeze dried from mixed solution of chitosan and type I collagen.Digital moisture monitor for skin was used to measure its moisture retention on rats.And its antibacterial property against Staphylococcus aureus and Escherichia coli was examined by impregnated paper disk method.CCK-8 method was used to measure its effect on the proliferation of mouse embryonic fibroblasts.Wound model was established on the back of rats.Set experimental group(treated with chitosan-collagen sponge),chitosan control group(treated with chitosan sponge)and collagen group(treated with collagen sponge)and observed wound healing process.Regenerative tissues were resected from the rats on day 3,7 and 14 after the establishment of wound models and the areas were recorded.Histopathological measurement was done after HE staining and Masson staining.ResultsThe chitosan-collagen sponge had little effect on cell proliferation of NIH-3t3 and exhibited inhibition against Staphylococcus aureus and Escherichia coli,and had a potent moisture retention.The wound area of experimental group was significantly smaller than chitosan control group(p<0.01)on day 3,7,14 and collagen group on day 3,7(p<0.05).The microscopic appearances of experimental group were better than chitosan control group and collagen control group.The time required for healing of wound was 14.38±1.06 days in experimental group,which was shorter than chitosan control group(17.00±0.82 days,p<0.05).No significant difference was observed between experimental group and collagen control group(15.25±1.28 days,p>0.05).ConclusionThe chitosan-collagen sponge has potent antibacterial property,moisture retention and promotive effect on wound healing.The chitosan-collagen sponge has no potential cytotoxicity.
