Objective: To observe the effect of MIF on salivary adenoid cystic carcinoma cell line(SACC)effect of transplantation tumor growth in nude mice,and to explore the ISO-1 inhibitors as anti-cancer drugs in clinical application.Methods: 40 nude mice with SACC-83 cell subcutaneous axillary transplantation were established.The nude mice were randomly divided into 10 groups(5 males and 5 females)according to the size of the transplanted tumors.The dosage of the drug was determined according to previous studies in this group.Namely,saline group,ISO-1 200 umol/L group,ISO-1 100 umol/L group,rMIF 50ug/L group,rMIF 25ug/L group,8 in each group,4 in male and 4 in female,were fed in separate cages.After measuring the volume of transplanted tumors in each group,there was no statistical significance.Each 0.2 ml tail vein was administered once every other day for 15 times.The length and short diameter of the transplanted tumors were measured before each injection,and the tumorigenesis curve was drawn.After 30 days of administration,the nude mice were executed,the tumors were taken out and weighed,and the inhibition rate was calculated [(1-the average weight of tumors in the experimental group/the average weight of tumors in the control group)*100%].Brain,lung and tumor tissues of nude mice were fixed,paraffin embedded and HE stained.Results: 1.The model of SACC subcutaneous transplantation in nude mice was successfully constructed.2.Compared with the control group,the volume growth of transplanted tumors in the ISO-1 concentration group was significantly smaller than that in the control group(P < 0.05).There was a significant difference in the anti-tumor rate between ISO-1 200 umol/L group and ISO-1 100 umol/L group(P < 0.05).The inhibition rate of ISO-1 200 umol/L group was 49.6%,and that of ISO-1 100 umol/L group was 40.94%.2.All nude mice were killed after 30 days of drug intervention.No metastasis was found in the lung and brain tissues of the control and experimental groups.Conclusion: 1.MIF can promote the growth of SACC Balb/c xenografts in nude mice,while ISO-1 can effectively inhibit the growth of SACC Balb/c xenografts in nude mice.ISO-1,200 umol/L group had more obvious inhibitory effect on transplanted adenoid cystic carcinoma of human salivary gland in nude mice than 100 umol/L group.2.Brain and lung metastasis were not observed 30 days after successful implantation.The conclusion is that the time for implantation of transplanted tumors is not enough.
