Meningitis is a common infectious disease of the central nervous system.The main types of meningitis are tuberculous meningitis,suppurative meningitis and viral meningitis.The prognosis of purulent meningitis and tuberculous meningitis is poor,which needs early diagnosis and timely and reasonable treatment.However,there is a lack of specific clinical manifestations in the early stage of meningitis,and there are no typical features of cerebrospinal fluid biochemical indicators and imaging findings.The basis for the diagnosis of neuroinfectious diseases lies in the detection of pathogenic organisms from the brain parenchyma or cerebrospinal fluid,but the traditional detection method of pathogenic microorganisms takes a long time,and the positive rate poor,so the classification of early meningitis is very difficult.Therefore,there is a necessary to find a rapid and specific method for the diagnosis of meningitis.Objective: To identify the protein with m/z=2176.205 and m/z=3399.302 in the classificatory diagnosis tree of meningitis which has established using surface-enhanced laser desorption ionization time-of-flight mass spectrometry(SELDI-TOF-MS)by liquid chromatography-tandem mass spectrometry(liquid chromatography-tandem mass spectrometry,LC-MS/MS).Methods: 62 cases of cerebrospinal fluid(CSF)with high contents of m/z=2176.205 and m/z=3399.302 were selected and detected by liquid chromatography-double pressure linear ion trap high resolution combined mass spectrometer(LC-LTQ Orbitrap Velos).The data was visualized by MZmine v2.27 and the protein was identified by Mascot v2.3.02.Then the obtained proteins were verified by enzyme-linked immunosorbent assay(Elisa).Results: M/z=2176.205 was identified as ubiquitin carboxyl terminal hydrolase 1(UCHL1)by liquid chromatography-tandem mass spectrometry(LC-MS)and m/z=3399.302 was identified as macrophage mobility factor(MIF).But,m/z=3399.302 is supported as MIF and m/z=2176.205 is not supported as UCHL1 by Elisa.Conclusion: The protein with m/z=3399.302 is MIF,and increases in patients with suppurative meningitis,which perfects the diagnostic tree of meningitis classification.
