In recent years,the incidence of gastric ulcer disease has gradually increased,and it was easy to recurrent.According to the clinical application of traditional Chinese medicine in treating gastric diseases,it was found that ginger licorice had a good effect,but at present,the most widely used in the market was baked licorice with honey,and the processing technology and efficacy of ginger licorice were less studied.Objective: In order to ensure the smooth development of the experiment and the reliability and stability of the experimental results,the quality of a large number of licorice medicinal materials was investigated and the high-quality medicinal herbs were selected as the research object,besides,the ginger juice was extracted as the auxiliary material.Under the guidance of the theory of traditional Chinese medicine,the optimal processing technology of ginger licorice was optimized,then the preventive and therapeutic effects of ginger-licorice on the rat gastric mucosal injury induced by restrained cold water stimulation and its stress mechanism were explored.Methods:In the research,the quality evaluation of licorice medicinal materials collected,mainly including the determination of water,ash,active ingredient content and fingerprint of the medicinal materials.In addition,the identification of the medicinal materials,heavy metal content and pesticide residues were also detected.Secondly,according to the quality evaluation results,a batch of high-quality medicinal herbs were selected,and the selected were used as the processing object,and ginger juice as the auxiliary material to study the optimal processing technology of ginger licorice;finally,the rat model of gastric mucosal injury induced by restrained cold water stimulation was establishedl.According to gastric ulcer index and ulcer inhibition rate,H&E staining,stress-related biochemical indicators of NO,MDA,SOD,GSH-Px,the pathological changes of gastric tissue were detected and the preventive effects of ginger licorice on gastric mucosal injury were analyzed,and the content of PGE2 in serum and tissue and pepsin activity in gastric juice were determined.Results:According to the determination of water,ash,glycyrrhizin,glycyrrhizic acid and fingerprint data of the original medicinal materials,a batch of high-quality licorice medicinal materials were selected.In the preparation process of ginger licorice,the appearance of ginger licorice and the content of main flavonoids were comprehensively evaluated to determine the optimum processing technology.Take licorice slices of each serving 100 g,add 50 mL ginger juice,mix well,suffocate 60 min and place in a container(fried medicine pot),stir fry at 60 ° C for 40 min,remove and let cool,sieve to remove debris.The rat model of gastric mucosal injury induced by restrained cold water stimulation was established.After the establishment of the model,the rats would suffer from loss of appetite and mental retardation and be found gastric bleeding after taking gastric anatomy.After the model is treated with drugs,it was found that licorice,ginger juice,ginger-licorice and honey-licorice had different preventive and therapeutic effects on gastric ulcer rats.The pathological section results clearly show the difference of gastric pathological morphology between model group and normal group.The results of NO,MDA,SOD,GSH-Px and pepsin activity showed that medicated group could have significant regulation effects on various biochemical indexes(p<0.01).In addition,the content of PGE2 in serum and stomach tissues was detected,and the effect of honey on the content of PGE2 in gastric tissues was not statistically different.Conclusion:Under the guidance of the technical theory of traditional Chinese medicine(TCM),this paper selected a batch of high-quality licorice medicinal herbs as the research object,and established optimal processing technology of ginger licorice.The preventive and therapeutic effects of ginger-licorice on the rat gastric mucosal injury disease induced by restrained cold stimulation were studied.It was verified by systematic experimental data that ginger-licorice had a good effect on preventing gastric mucosal injury.This study provided scientific basis for clinical use of traditional Chinese medicine.
