Purposes:1.To investigate the diagnostic value of digital subtraction angiography(DSA)perfusion imaging parameters PBV in rabbit liver VX2 tumors and its correlation with CT perfusion imaging parameters.2.To investigate the ability of digital subtraction angiography(DSA)perfusion imaging parameters PBV to evaluate the embolization effect of VX2 liver tumor in rabbits.Materials and methods:Twenty-three New Zealand white rabbits,male or female,weighing 2.0-3.Okg,were implanted with 0.2-0.3ml tumor tissue in the left lobe of the liver of each rabbit under CT-guidance.No-enhanced and enhanced CT scanning confirmed that 20 of the 23 rabbits had a single lesion in the left lobe of the liver,which met the experimental criteria.Twenty rabbits with VX2 hepatocellular carcinoma were randomly grouped into 8 rabbits in group A and 12 rabbits in group B.In the VX2 liver cancer model of group A,CT perfusion imaging was performed first,and the perfusion parameters were obtained using post-treatment software,including maximum intensity projection(MIP),blood flow(BF),blood volume(BV),arterial liver perfusion(ALP),portal liver perfusion(portal liver perfusion).PVP)hepatic perfusion index(HPI),then,DSA perfusion imaging parameters(PBV)were measured by hepatic artery catheterization,and consistency between CT perfusion parameters and DSA perfusion parameters are compared in tumor active area and normal liver tissues around tumor.After the experiment,the animals were sacrificed and the expression of microvascular density in the tumor tissue(microvascular density,MVD)and vascular endothelial growth factor(vascular endothelial growth,VEGF)was detected by immunohistochemical method to compare with DSA perfusion parameters PBV correlation analysis.The DSA perfusion imaging parameters PBV were obtained by hepatic artery intubation in group B,then TAE embolization,and the DSA perfusion imaging parameters PBV were obtained by hepatic artery intubation last.The changes of DSA perfusion parameters(PBV)were calculated before and after embolization.The experimental animals were sacrificed on the 7th day after embolization to observe the necrosis of the tumor tissue,and the expression of MVD and vascular endothelial growth factor in tissues of the tumor group was detected by immunohistochemical method.The correlation between ?PBV and MVD and VEGF in tumor tissues after embolization was compared,and receiver operating characteristic curve(ROC)was used to determine the optimal threshold for thorough embolization effect diagnosis of tumor tissue DSA perfusion parameter PBV change rate(?PBV%)before and after embolization.Results:1.Scatter plots were drawn and fitted for the CT perfusion imaging tumor diameter measurements and DSA perfusion imaging tumor diameter measurements in 8 tumor rabbits in group A,with A high degree of consistency(R2=0.833).2.The correlation coefficients of DSA perfusion parameters PBV and CT perfusion imaging parameters MIP,BF,BV,ALP,PVP and HPI in tumor tissues were 0.647,0.611,0.838,0.922,0.515 and 0.898,respectively,and the P values were 0.083,0.007,0.009,0.006,0.192 and 0.102,respectively.Tumor PBV was significantly correlated with BF,BV and ALP,and all of them showed positive correlation.3.The correlation coefficient between the pathological parameters of VEGF and MVD was 0.845,P=0.008<0.05.There was a significant correlation between MVD and MVD,showing a positive correlation.4.Tumor PBV DSA perfusion parameters and normal liver tissue around the tumor CT perfusion parameters of normal liver tissue around MIP,BF,BV,ALP,PVP,HPI correlation coefficient respectively:0.148,0.636,0.946,0.764,0,0,P values were 0.727,0.008,<0.0001,0.028,1,1,tumor around PBV normal liver tissues and normal liver tissues surrounding tumor BF,BV,ALP has significant correlation,and there was a positive correlation.5.The correlation coefficients of DSA perfusion parameters PBV and pathological parameters VEGF and MVD in tumor tissues were 0.850 and 0.802,respectively,and the P values were 0.007 and 0.017,respectively.PBV in tumor tissues was significantly correlated with VEGF and MVD,and both showed positive correlation.6.PBV of tumor tissue before TAE was 50.33 8.25 ml/L,and that after TAE was 13.25 10.27 ml/L.PBV of tumor tissue after TAE was significantly lower than that before TAE,and the difference was statistically significant(P<0.05).7.Correlation coefficients of VEGF and MVD were-0.759 and-0.760,respectively,and P values were 0.004 and 0.004,respectively.Significant correlation and negative correlation existed between PBV and MVD and VEGF.8.The area under ROC curve was 1.000 and the diagnostic threshold was 58.3%in the group with complete TAE embolization effect(?PBV%).conclusions:1.Both DSA perfusion imaging parameters and CT perfusion imaging can be used to understand the local blood supply characteristics of VX2 liver cancer lesions in rabbits by calculating the blood perfusion volume of hepatic artery.There is a positive correlation between DSA perfusion imaging parameters PBV and CT perfusion imaging parameters BV,BF and ALP.2.DSA perfusion parameter PBV can be used to evaluate the residual blood supply of VX2 liver cancer in rabbits after TAE,and a positive correlation between PBV and embolization effect can be found;PBV%>58.3%indicates complete embolization effect of VX2 liver tumor in rabbits.
