Objective: 1.Firstly,the gastric cancer chip was retrieved through the GEO database,and then the differentially expressed genes were screened using the GEO2 R online data analysis tool.The long noncoding RNAs with obvious differential expression were selected,and the research subjects were determined according to the literature.2.To analyze the expression level of linc RNA-p21 in gastric cancer tissues and adjacent normal tissues and its relationship with clinical data of patients with gastric cancer.To explore the role of linc RNA-p21 in the occurrence and development of gastric cancer,and provide a new direction for the diagnosis and treatment of gastric cancer.Methods: 1.Go to the GEO database home page and search for gastric cancer related chips.Find the gene chip data of interest in the search results,and comprehensively compare and analyze to determine the gastric cancer gene chip.Then click on "Analyze with GEO2R" below.Firstly,the basic data distribution of the samples in the chip is verified.If the chip sample data is uniform,the samples are grouped.The intra-chip samples were divided into gastric cancer tissue group and paracancerous tissue group,and the GEO2 R online data analysis tool was used for data processing.The GEO2 R analysis found differentially expressed genes and selected lnc RNAs with significant differential expression.Finally,by reading the relevant literature and other comprehensive analysis and processing,a significant differential expression of lnc RNA was selected.2.Gastric cancer tissues and corresponding normal tissues adjacent to the cancer were collected from 58 patients with gastric cancer who were treated with gastrointestinal surgery from Qingdao University Affiliated Hospital from January 2018 to June 2018.And record the clinical and pathological data of each patient who meets the requirements.The relative expression level of linc RNA-p21 in gastric cancer tissues and adjacent normal tissues was detected by Quantitative Real-time PCR.The statistical analysis was used to analyze the differential expression of linc RNA-p21 in gastric cancer tissues and adjacent normal tissues,and further analyze the relationship between the relative expression level of linc RNA-p21 and clinical data of gastric cancer tissues.Results: 1.This study selected the gastric cancer gene chip GSE19826 in the GEO database.The homogeneity of the chip GSE19826 was verified by using the GEO2R online data analysis tool.The samples in the chip were then divided into a gastric cancer tissue group(12 cases)and a paracancerous tissue group(12 cases).The chip data was analyzed to obtain differential genes,and lnc RNAs with obvious expression differences were selected.There were 20 lnc RNAs with distinct expression differences,including 6 up-regulated lnc RNAs and 14 down-regulated lnc RNAs.By further reviewing the literature and other comprehensive screening,linc RNA-p21 was selected as the research object.2.The relative expression level of linc RNA-p21 in 58 cases of gastric cancer was detected by q PCR method was 0.27±0.12,which was significantly lower than in adjacent normal tissues(t=33.33,P<0.001).Statistical analysis showed that there was no significant correlation between the relative expression of linc RNA-p21 in gastric cancer tissues and the age,sex,tumor size,gross tumor type,preoperative BMI,preoperative CA125 level,preoperative AFP level,preoperative hemoglobin level and preoperative albumin level of gastric cancer patients(P>0.05).The levels of linc RNA-p21 in gastric cancer tissues were related with tumor location,differentiation degree,invasion depth,lymph node metastasis,TNM stage and preoperative CA199,preoperative CEA(P<0.05).Conclusion: The gene chip was selected through the GEO database,and the differentially expressed gene linc RNA-p21 was screened by GEO2 R online analysis tool.To verify the relative low expression of linc RNA-p21 in gastric cancer tissues,and further analyze the relationship between linc RNA-p21 and clinical data of gastric cancer patients.It is proved that linc RNA-p21 is closely related to the occurrence and development of gastric cancer,and may be a potential auxiliary diagnostic marker and a potential target for drug therapy.
