Effects Of 5 Medicines On Immune Response Of HPV Infected Mouse

[Objective] Currently used in clinical practice to prevent recurrence of condyloma acuminata are imiquimod,interferon and PaiTeLing.In this study,an animal model of HPV infection was established to study the immuno-stimulation effect of PaiTeLing,imiquimod,IFN-?-2b,SanBai lotion,and ChuYou on HPV infected mice.[Methods] In this study,we implanted the patient's warts(secretion of HPV types for HPV6,HPV31,HPV81,informed consent,patients with size of 0.5 cm ^ 3)under the right back of nude mice,successfully constructed a condyloma acuminatum animal model,and used the MSD kit to detect expression levels of IFN?,TNF?,IL-2 and IL-12 in the serum taken from nude mice's orbital venous and orbital artery,and HE staining and immunohistochemical analysis of the skin tissue at the mouse inoculation site.RNA was extracted from the warts using a total RNA extraction kit,and reverse transcribed into DNA for multiplex quantitative PCR to detect viral load.[Results] Compared with the negative control group,the levels of TNF?,IL-2,IFN-?and IL-12p70 were significantly increased in the treatment group;in HE and immunohistochemistry,koilocytes in the treatment group were significantly lower than the untreated group,the number of Langerhans cells increased,the positive expression of CD207 was more significant,and the HPV viral load of the treatment group also decreased significantly.In general,the imiquimod group and the PaiTeLing group showed a more obvious trend than other groups in these results.[Conclusion] Imiquimod,IFN-?-2b,PaiTeLing,SanBai and ChuYou can effectively treat condyloma acuminatum and enhance the immune response,among which Imiquimod and Paiteling are better than other medicines.
