| Objective:Ulcerative colitis is a kind of inflammatory bowel disease that is difficult to heal and has a high recurrence rate,which seriously affects the mental and psychological life of patients.In this experiment,we discussed the Mechanism of HuazhuoqingJieYukuiJian in the treatment of Ulcerative Colitis based on the turbidity theory to provide experimental basis for clinical treatment of UC and the role of probiotics in treating ulcerative colitis by observing the effects of HuazhuoqingJieYukuiJian on the general situation,disease activity index,pathological changes of colonic mucosa,the level of TNF-alpha,interleukin-17 in serum and the expression of cell adhesion factor-1 in ulcerative colitis model rats.Methods:60 CL male Wistar rats were randomly divided into six groups on average: group A(blank group),group B1(model group),group B2(traditional Chinese medicine group),group B3(western medicine group),group B4(Chinese-western medicine group)and group C(probiotics group).In addition to the blank group,the rat model of UC were established by using the method of combined use of 2,4,6-trini-trobenzene esulfonic acid /ethanol on the other groups.After successful modeling,rats in all groups ate freely.Rats in the model group were fed with normal saline.The traditional Chinese medicine group was treated with HuazhuoqingJieYukuiJian.The western medicine group was given mesalazine enteric-coated tablets.The traditional Chinese-western medicine group was give HuazhuoqingJieYukuiJian and mesalazine enteric-coated tablets.The probiotics group was fed with Bifidobacterium triple living bacteria capsule.At the end of the last administration,the TNF-α in serum was measured by enzyme linked immunosorbent assay(Elisa).The content of IL-17 and the expression of ICAM-1 in colonic mucosa by Immunohistochemistry.Results:1.General state and DAI score of rats: The general status of group A rats was normal,and DAI score was the lowest;The general state of B1 group was poor,and DAI score was the highest.Compared with other groups,the difference was statistically significant;The general status of rats in group B2,B3 and C was similar,and there was no statistically significant difference in DAI score.Group B2 was the lowest among the three groups;The general status and DAI scores of rats in group B4 were better than those in group B2,group B3 and group C,and the differences were statistically significan.2.Macroscopic and microscopic observation of colon tissueColonic tissue of group A rats was normal under naked eye and microscope.Macroscopically,rats in group B1 showed obvious hyperemia,edema and ulceration,with prominent microscopic destruction of glands and infiltration of inflammatory cells.The mucosal surface of group B2,group B3 and group C was slightly less smooth,with a small amount of hyperemia,edema,erosion and other changes.Microscopic observation showed that the glandular structure was slightly less orderly,with a small amount of inflammatory cell infiltration.The mucosal surface of rats in the B4 group was smooth,with occasional superficial ulcers.Microscopic observation showed clear and complete glandular structure,without obvious inflammatory cell infiltration.3.Expression of IL-17,TNF-α and ICAM-1IL-17 levels : Group A was the lowest,but there was no significant difference from group B4.There was no significant difference between group B2,group B3 and group C.The difference between group B4 and group B2,B3 and C was statistically significant.TNF-α levels: Group A was the lowest,but there was no significant difference from group B4.There was no significant difference between group B2,group B3 and group C.The difference between group B4 and group B2,B3 and C was statistically significant.The expression of ICAM-1: Group B1 had the highest expression level.Both group A and group B4 were lower than group B2,group B3 and group C,and the difference was statistically significant.There was no significant difference between group B2,group B3 and group C.Conclusion:1.HuazhuoqingJieYukuiJian had good effect on UC rats.The mechanism of HuazhuoqingJieYukuiJian in the treatment of UC may be that it can inhibit the inflammatory reaction by reducing the level of IL-17,TNF-α in serum,and down-regulate the expression of ICAM-1 to repair the damaged mucosa.The efficacy of group B2,group B3 and groupC was basically the same,but the reduction was more obvious in group B2.The combined effect of Chinese and western medicine is better than that of single drug.2.Bifidobacterium triad had therapeutic effect on UC rats.The mechanism of bifidobacterium trifecta in the treatment of UC may play a role by regulating the imbalance of intestinal flora,repairing the damaged intestinal mucosal barrier,and affecting the level of inflammatory factors. |
