Objective:By observing the effects of Huazhuojieduyukui Decoction on the general condition,disease activity index(DAI)score,histopathological changes of colon,IL-2 content in serum and ICAM-1 expression in colon tissue of ulcerative colitis(UC)model rats,discuss the mechanism of this decoction in curing UC.Method:60 rats were randomly divided into 6 groups,10 rats in each group: normal group(N group),model group(M group),mesalazine group(MS group),high dose of Huazhuojieduyukui decoction group(ZH group),medium dose group(ZM group)and low dose group(ZL group).All rats except the normal group,were built for UC animal model by using TNBS/ethanol combination method.4 days after successful modeling,the M group and other treatment groups were given normal saline and corresponding drugs respectively for 2 weeks.The general state of rats in each group was observed,and the DAI scores were performed,after the treatment,dissecting colon tissue to make pathological observation,taking the blood to detect the content change of IL-2 and measuring the expression of ICAM-1.Results:1.General stateCompared with N group,the general state of rats in other groups became worse after modeling.After 2 weeks treatment,the general state of ZH group is similar to that of N group;the improvement of ZM and MS group showed no significant difference,the improvement of these two groups was better than that of ZL and worse than ZH group;the M group was worse than ZL group.2.DAI score resultsZH,ZM,ZL and MS groups were lower than M groups;the ZH group was lower than the ZM,ZL and MS groups;ZM and MS group were lowerthan ZL group;All the differences mentioned above have statistical significance.(P<0.05).There was no significant difference between ZM group and MS group.(P>0.05).3.Macroscopic and pathological observations of colon tissueThe N group was normal;the most serious mucosal damage was found in M group;the ZH group was close to the N group;ZM group was similar to MS group,these two groups were inferior than ZH group and better than ZL group;ZL group was better than M group.4.Index detectionThe content of IL-2: the content of IL-2 in N group was higher than that of other groups;ZH group was higher than ZM,ZL,MS and M group;ZM group and MS group were higher than those in ZL and M group;ZL group was higher than M group;all the above differences were statistically significant(P<0.05).There was no significant difference between ZM and MS group(P>0.05).The expression of ICAM-1: compared with other groups,N group had the lowest expression;ZH group was lower than ZM,ZL,MS and M group;the ZM and MS group were lower than ZL and M group;ZL was lower than M group;all the above comparisons were statistically significant(P<0.05).There was no significant difference between ZM and MS group(P>0.05).Conclusion: Huazhuojieduyukui decoction can effectively improve the general situation of the UC rats,reduce colonic mucosa injury and promote repair,and treatment effect is related to the dosage of this decoction.Its mechanism may be through increasing the content of IL-2,reducing intestinal tissue ICAM-1 expression,alleviate inflammation,regulate immunity and promote intestinal mucosal healing.
