Objective To investigate whether there is a correlation between estrogen and the gene polymorphism of estrogen receptor ER?(rs2077647,rs3798758)with hypertension in the elder group(age?60)of urban communities of Ningxia and explore the relationship between estrogen levels and hypertension in the elder group of urban communities of Ningxia,the frequency distribution of estrogen receptors,and to study the correlation between gene polymorphism and the incidence of hypertension on a molecular basis.Methods The elder groups in five communities(Qinghe District,Xianghe District,etc.)of Ningxia,Ningxia were selected and investigated by cluster stratified sampling.There were a total of 1263 people involved in the study with ages over or equal to60,including 608 males and 655 females,967 of Han nationality and 296 of Hui nationality,of which the average age was 69.80 for males and 67.60 for females.The sample genotyping technique MassARRAY system was used to record and the statistical software was used to analyze the relationship between estrogen levels and genotypes with blood pressure in different populations in the communities.Results 1.In the elder male group,the testosterone level decreased with age increase,and the difference among groups was statistically different(P<0.05).Systolic pressure increased gradually and the difference was statistically significant(P<0.05).Diastolic pressure decreased first and then increased slightly and the difference was statistically significant(P<0.05),but it showed a downward trend overall.Estradiol increased first andthen decreased and the results were statistically different(P<0.05),but it showed an downward trend overall.2.In the elder female group,diastolic pressure decreased with age increase and there was statistical difference among different age groups(P<0.05),overall,it showed a downward trend.Systolic pressure gradually increased,and the difference was statistically significant(P<0.05),overall,it showed an upward trend.Estradiol increased first and then decreased,and the difference was statistically significant(P<0.05).3.In elder female group,there was statistical significance in the distribution of estrogen levels in hypertensive group and normal group(P<0.05).Correlation analysis in female groups suggests that estrogen is associated with hypertension in females.4.The genotype distribution frequency was tested by Hardy-Weinberg genetic balance(HWE),which showed it was in line with genetic balance and had genetic representativeness for people in this study.5.In this study,the genotype distribution frequencies were different among elder groups of Han nationality under allele and dominance models(AA+CA)of locus rs3798758.6.Compared with the different genomic systolic and diastolic blood pressure levels of the two sites,the rs3798758 locus in the dominant model showed that the average level of diastolic blood pressure was lower in individuals carrying AA or CA genotype than those carrying CC genotype,and the difference was statistically significant(P <0.05).7.(1)Risk assessment on the prevalence rate of hypertension of different genotypes of locus rs2077647 was conducted in different ethnic groups and gender groups,and individuals carrying C mutations in females and males of Han nationality groups may have a reduced risk of hypertension;(2)In the males of Han nationality groups of this study,the distributions of the prevalence rate of hypertension of different genotypes(co-dominance model: CC/CA/AA,dominance model:AA+CA/CC)of rs3798758 were different(P<0.05),and individuals carrying A mutations may have an increased risk of hypertension.Conclusions There do exist a correlation between estrogen level and polymorphism of the locus gene in this study with hypertension in different genders and ethnic groups of theelder in urban communities of Ningxia.
