Experimental Study On Anti-tumor Effect Of Liquor-saturated Stellera Chamaejasme L. On S180 Tumor-bearing Mice

Objective:Through the establishment of S180 subcutaneous tumor-bearing mouse model,the effects of wine on the anti-tumor activity and toxicity of Stellera chamaejasme L.were studied,and the anti-tumor mechanism was preliminarily explored.Methods:80 Kunming mice were randomly selected 10 mice as blank controls.The 70 remaining were in the left upper extremity of the mouse,and the S180 sarcoma was subcutaneously modeled.The remaining 70 were subcutaneous tumor-bearing models of S180 sarcoma in the left upper extremity of the mouse.Tumor-bearing mice were randomly divided into 7 experimental groups: model group,low-medium-high dose group of crude drug of stellera chamaejasme and low-medium-high dose group of liquor-saturated stellera chamaejasme.Taking a mode of administration by gavage,continuous medication for 7 days.The blank control group and the model group were given physiological saline;according to their body weight,the crude drug group and the liquor-saturated group were given a concentration 5mg/kg,10mg/kg,20mg/kg of decoction of crude drug or liquor-saturated of stellera chamaejasme.After 24 hours of the last administration,the mice were weighed and execution of experimental animals by cervical spondylolisthesis.Dissecting mice,tumor tissues,spleen,thymus were extracted and weighed.The microstructure of tumor tissue,spleen and thymus was observed by HE staining.The expression levels of Ki-67 and caspase-3 in tumor tissues were detected by immunohistochemistry.Results :1.The tumor tissue weight of the mice in the drug group and the liquor-saturated group were lower than that of the model group(P<0.05).The tumor inhibition rates of the low,medium and high dose groups of crude drugs were respectively 34.80%,44.00% and 33.18;The anti-tumor rate of the low,medium and high dose group of liquor-saturated group were respectively 46.80%,57.97% and 54.95%.The tumor tissue growth can be inhibited by the groups of crude drug and liquor-saturated.The anti-tumor effect of liquor-saturated group was better than that of crude drug group(P<0.05).Among them,the liquor-saturated medium dose groupwas the highest(57.97%).2.Compared with the blank control group,no significant changes of spleen index occurred in the the liquor-saturated of Stellera chamaejasme;the spleen index of the crude drug groups were decreased,among which the decrease was most significant in high dose group(P<0.05).There was no significant change in thymus index in each group(P>0.05),spleen and thymus did not have significant morphological changes.3..Morphological observation showed that compared with the model group,the number of tumor cells in each dosage group of crude drugs and wine was less and the arrangement was sparse.Most of them were scattered in flaky light pink necrosis area.Normal tissue structure disappeared in necrosis area,cell membrane ruptured,nucleus fragmented or disappeared.Leydig cells increased and leukocytes infiltrated in different degrees.4.Immunohistochemistry test results show: Stellera chamaejasme liquor-saturated can inhibit the expression of ki-67.Among them,the high-dose group of wine had the lowest proliferation index of Ki-67,which was 26.50%±8.90% and 26.28%±7.23%,respectively.Stellera chamaejasme liquor-saturated can promote the expression of caspase-3 in tumor tissues of S180 tumor-bearing mice(P<0.01,P<0.05).Among them,the high-dose group in the wine system had the highest positive expression rate of Caspase-3,which was 50.75%±18.48% and 40.40%±17.60%,respectively.Conclusion:1.Liquor-saturated can enhance the inhibitory effect of stellera chamaejasme on S180 subcutaneous tumor-bearing mice.2.In addition to enhancing the anti-tumor effect,liquor-saturated can reduce the effects of Stellera chamaejasme on body weight and spleen index of mice.3.By down-regulating the expression of ki-67,Stellera chamaejasme liquor-saturated can inhibit the proliferation of tumor cells in S180 subcutaneous tumor-bearing mice.By up-regulating the expression of caspase-3,Stellera chamaejasme liquor-saturated can promote the apoptosis of tumor cells in S180 subcutaneous tumor-bearing mice.
