Objective: With the increasing incidence of ulcerative colitis,it is extremely urgent to find an effective treatment method.By consulting the literature,we selected the Shaoyao Decoction with clearing heat and dampness effect to observe the rats with moist heat-enriched ulcerative colitis.The effects of IRAK-1,TRAF-6,and IL-8 in colon tissue explored the relationship between TLR4/NF-κB signaling pathway and damp-heated ulcerative colitis and the effect of Shaoyaotang on this pathway.Methods The humic-thermal intensive UC rats were duplicated by high-fat,high-sugar spicy diet plus immune complex method + TNBS combined with ethanol.They were randomly divided into blank group,model group,Shaoyaotang high,medium and low dose group,and sulfasalazine group.10 males and half females in each group were sacrificed on the 21 st day after drug intervention.The colonic lesions were taken from the rats.Real-time PCR was used to detect m RNA and Western blot was used to detect protein expression.,pathological sections were observed by Su Yihong-wood essence(HE)staining,and m RNA and protein expression of IRAK-1,TRAF-6 and IL-8 in colon tissues were detected.And observe the uncle disease activity index,mucosal injury and pathological score of rat colon tissue.result 1.Activity and status Through the general morphological observation of the rats,the symptoms of each treatment group were improved.The symptoms of the high dose group of Shaoyao Decoction and the sulfasalazine group were the most obvious.The high-dose group of Shaoyao Decoction had the same degree of symptom relief as the SASP group.The DAI score was statistically significant(P < 0.05).From the CMDI score of rat colon tissue,compared with the blank group,the CMDI score of the model group was significantly increased(P<0.05).Compared with the model group,the CMDI scores of each treatment group were significantly decreased(P<0.05),among which high dose The group is most obvious2.Colon tissue pathology Most of the colonic tissues of the model group penetrated into the muscular layer,and the lesions were severe.The treatment groups changed to different degrees,the mucosa began to repair,scar tissue formed,and new glands were seen.The histopathological score of colon tissue in UC rats was significantly lower than that in the model group(p<0.05).3.Expression of m RNA and protein in colon tissue Compared with the blank group,the expression of IRAK-1,TRAF-6,IL-8 protein and m RNA in the model group was significantly increased(P<0.05).Compared with the model group,each group of Shaoyao Decoction and sulfasalazine group IRAK-1 The expressions of TRAF-6,IL-8 protein and m RNA were decreased in different degrees,and the most significant doses of Shaoyao Decoction and sulfasalazine were the most significant(P<0.05).conclusion: 1.The occurrence of UC has a certain correlation with the TLR4/NF-κB pathway,which may be one of the mechanisms of UC pathogenesis.2.Shaoyao Decoction can inhibit the key transduction molecules IRAK-1 and TRAF-6 in the TLR4/NF-κB signaling pathway of damp-heat UC rats,thereby inhibiting the signal transduction of TLR4/NF-κB signaling pathway and reducing the release of inflammatory factors.Thereby,the inflammation of the moist heat-growth type UC rat is alleviated.
