In this paper,Salvia miltiorrhiza Bunge was used as the research object to explore the influencing factors of the stability of the active ingredients of Chinese herbal medicines and the measures to stabilize the herbs.That is,the S.Bunge medicinal materials are first treated differently(light and dark,different temperature treatment,Co~600 irradiation,S.Bunge is used in the compatibility of common medicinal materials)to clarify the main factors affecting the content of the active ingredient tanshinone IIA;on this basis,the raw materials are processed.The?-cyclodextrin inclusion treatment was designed by Box-Behnke response surface test.The encapsulation efficiency of tanshinone IIA and free amino acid was used to evaluate the preparation process of inclusion complex.The retention of tanshinone IIA in light,temperature and compatibility of S.Bunge-?-cyclodextrin and Pheretima-?-cyclodextrin inclusion complex was detected by scanning electron microscopy(SEM)and Fourier transform infrared spectroscopy.(FT-IR)and differential scanning calorimetry(DCS)were used to identify the surface morpHology,functional group changes and thermal stability of the inclusion complex.The human intestinal juice environment was further simulated,and the in vitro release of the inclusion complex was investigated.The following main results have been achieved:1.Light and Co~600 irradiation had no significant effect on the content of tanshinone IIA in S.Bunge.Heating could promote the content of tanshinone IIA.The content of tanshinone IIA was not significantly decreased after the combination of medicinal herbs and S.Bunge.However,the compatibility of animal herbs with S.Bunge can cause tanshinone IIA content.Significantly reduced(40%reduction in tanshinone IIA retention under the conditions of thisresearch).2.Using?-cyclodextrin to encapsulate S.Bunge medicinal herbs by saturated aqueous solution method can effectively alleviate the degradation of tanshinone IIA in Salvia miltiorrhiza.The preparation process of S.Bunge-?-cyclodextrin inclusion compound after response surface optimization was as follows:inclusion time 4 h,inclusion temperature50?,mass ratio of?-cyclodextrin and salvia miltiorrhiza was 1:1,encapsulation of S.The rate was 40.74%;the?-cyclodextrin was used to encapsulate the animal medicinal material(Pheretima),and the preparation process of the inclusion compound was as follows:inclusion time 4 h,inclusion temperature 50°C,?-cyclodextrin and earthworm When the mass ratio is 1:1,the enveloping rate of the earth P.reaches 91.64%.The formed inclusion complex was observed by scanning electron microscopy.The inclusions of S.were arranged in disorder and closely connected,which was a smooth elliptical shape.The crystal form of Pheretima ainclusions showed a spherical phase.Infrared spectroscopy revealed The migration and peak intensity of the characteristic peaks before and after inclusion were observed.Differential scanning calorimetry examined the difference in thermal stability of the materials before and after inclusion,indicating that the drug was successfully encapsulated or adsorbed into?-cyclodextrin.3.Light and temperature had significant effects on the stability of S.Bunge-?-cyclodextrin inclusion complex.Continuous light caused the decomposition of?-cyclodextrin,which resulted in no significant content of tanshinone IIA in S.Bunge inclusion compound and unincorporated S.Bunge difference.The S.Bunge-?-cyclodextrin inclusion complex was significantly protected from the retention of tanshinone IIA in the dark,and the retention was 95%during the experimental period.After the combination of S.Bunge inclusion compound and common medicinal materials,the retention of tanshinone IIA was twice as high as that of non-inclusion.The content of tanshinone IIA was inhibited after the inclusion of Pheretima.Under the accelerated test conditions of 65°C,the retention of tanshinone IIA in the mixture of Pheretima-?-cyclodextrin inclusion compound and S.Bunge was about 30%higher than that of uncompounded compatibility.4.After?-cyclodextrin inclusion,the total in vitro cumulative release of total free amino acids and tanshinone IIA in S.Bunge and Pheretima was increased.The S.Bunge inclusion compound is 10%higher than the unencapsulated environment in the simulated human intestinal juice environment,and the earthworm inclusion compound is about 6%higher than the unencapsulated compound.
