Dan shen is the dry root of the origanum Salvia miltiorrhiza Bge. It is bitter intaste and cold-natured and has been used as a traditional Chinese folk medicine for a long time. It have been widely used in curing the disease of cardiovascular system clinically. At present, with the expanding of clinical use of DanShen, some researchers studied the stem and the leaf of it, proving that they have the similar bioactives and pharmacological actiong as the root.In order to exploit the resource of the aerial part of Salvia miltiorrhiza Bge., this paper carried out a systematic pretest on chemical constituents, distribution traits of bioactives in different parts of Salvia miltiorrhiza Bge. We also determinated the mono-salvianolic acid compounds of leaf.The results were as follows:1. Studied the chemical constitutions of the leaf and stem of Salvia miltiorrhiza Bge.. The results showed that there were phenols, tannin, organic acid, alkaloid, steroid, flavonoids,anthraquinone coumarin and terpenoidm in the two parts2. Routine analyses of leaf and stem displayed that the contents of amylase is 17.90%,13.27%, crude fat is 4.48%,3.03%,protein is 30.30%,24.5% respectively.3. The contents of total phenolics acid and total flavonoids of deferent parts of Salvia miltiorrhiza bunge. have been determined. And the Antioxidant activities in vitro of the extractions from root, leaves and stem of Salvia miltiorrhiza bunge. have been evaluated under the reference of vitamin C. The results indicated that the content of total phenolics acid and total flavonoids in the leaf is significantly higher than the root and stem. The total phenolics in the leaf was 1.07 times and 6.81 times of the root and the stem respectively. And the total flavonoids in the leaf was 4.79 times and 14.5 times of the root and the stem respectively. The results showed that the Antioxidant activities of ethanol extraction of leaf is significantly higher than the root and stem. And the Fe3+ to Fe2+ reducing activity, DPPH free radical scavenging activity and the inhibitory effect of PUFA was 1.08, 1.01 and 5.89 times of vitamin C(1mg/mL) respectively. A positive correlation was observed between total phenolic acid content and antioxidant activities. It demonstrated that the antioxidant activity may be in relation to the total phenolic acid content.4. Based on the basis of single-factor analysis, A 4-factor, 3-level Box-Behnken experimental design was applied and the interactions of each factor on the polyphenolic yield were also investigated. The optimal conditions were at 50.00℃extraction temperature, 33:1mL/g liguid-solid ratio, 63% alcohol concentration and 43 min extraction time. Under such conditions, the polyphenolic yield is 7.78% after three runs. The validation test shows that the prediction values of the model are well agreed with the experimental values.5. Purification of total phenolic acid crude extract from the leaf of Salvia miltiorrhiza bunge. .NKA-9 resin was chosen for the separation. The results revealed that the largest adsorption capacity was achiveved when initial phenolic concentration was 3.5mg/mL, pHwas 6, feed flow rate was 2.0mL/min and feed volume was 7BV.The saturated resin was first washed with 3BV of water to remove impurities,then desobed with 2.2BV of 75% alcohol at flow rate of 1.5mL/min.The desorption ration of total phenolic acid from resin was up to 93%,and the purity of total phenolic acid was increased to 51.8%.6. Seven mono-phenolic compounds in crude were determined by HPLC. The results indicted that seven mono-phenolic compounds were obtained from he leaf of Salvia miltiorrhiza bunge.. The contents of Ros is the highest ,it reached 88.03mg/g.The order of the contents as follows:Ros( 88034.95ug/g ) >Caffeic acid ( 761.00ug/g ) >SalB ( 414.95ug/g )>Danshensu( 342.45ug/g) >Ferulic acid( 283.59ug/g) >SalA( 144.48)ug/g>Protocatechualdehyde(113.49ug/).
