Objective:To detect the expression level of IncRNA AC104695.4 in bladder urothelial carcinoma and explore its clinical significance.Methods:The expression levels of IncRNA AC104695.4 in different bladder cancer cells(BIU87,EJ,RT4 and T24)and the normal bladder epithelial cell(SV-HUC-1)were detected by qRT-PCR.Then,the expression levels of IncRNA AC104695.4 in tumor tissues and corresponding adjacent tissues of 47 patients with bladder urothelial carcinoma were further detected by qRT-PCR.Next,detecting the expression levels of IncRNA AC 104695.4 in tumor tissues and corresponding adjacent tissues of other patients which include 13 cases of prostate cancer,12 cases of renal carcinoma,11 cases of adrenal tumor and 10 cases of ureteral cancer.In addition,agarose gel electrophoresis experiments were performed to clarify the specificity of the qRT-PCR products.At the same time,analyzing the correlation between the expression level of IncRNA AC 104695.4 and clinical parameters which include sex,age,recurrence,tumor number,tumor diameter,pathological grade,clinical stage,depth of invasion and lymph node metastasis in patients with bladder urothelial carcinoma.The ROC curve was used to further clarify the sensitivity and specificity of IncRNA AC 104695.4 in the diagnosis of bladder urothelial carcinoma,and then exploring the clinical significance of IncRNA AC104695.4 in the diagnosis of bladder urothelial carcinoma.Results:1.Compared with the normal bladder epithelial cell(SV-HUC-1),the relative expression levels of IncRNA AC 104695.4 in bladder cancer cells(BIU87,EJ,RT4 and T24)were(0.46±0.08),(0.11±0.07),(0.26±0.11)and(0.08±0.03).All of them were at a low level of expression,and the difference was statistically significant(F=88.75,p<0.05).2.The relative expression level of IncRNA AC104695.4 in bladder urothelial carcinoma tissues was(9.93±12.26),which was significantly lower than that in the corresponding adjacent tissues whose relative expression was(46.61±31.05),and the difference between them was statistically significant(z=-6.499,p<0.05).In 47 cases of bladder urothelial carcinoma,44 cases of tumor tissue showed low expression of IncRNA AC 104695.4,and the positive detection rate of bladder urothelial carcinoma was as high as 93.62%.3.There was no significant difference in the relative expression level of IncRNA AC104695.4 between prostate cancer tissues and corresponding adjacent tissues(z=-0.231,p=0.817>0.05).There was no significant difference in the relative expression level of IncRNA AC 104695.4 between renal carcinoma tissues and corresponding adjacent tissues(t=-0.473,p=0.641>0.05).There was no significant difference in the relative expression level of IncRNA AC 104695.4 between adrenal tumor tissues and corresponding adjacent tissues(z=-0.033,p=0.974>0.05).But the relative expression level of IncRNA AC 104695.4 in ureteral urothelial carcinoma tissues was(0.85±0.28),which was lower than that in the corresponding adjacent tissues whose relative expression was(2.02±0.92),and the difference between them was statistically significant(t=-3.846,p=0.003<0.01).4.There was no correlation between the expression level of IncRNA AC 104695.4 and gender,age,recurrence,tumor number,tumor diameter,lymph node metastasis and clinical stage in patients with bladder urothelial carcinoma(p>0.05).However,the expression level of IncRNA AC 104695.4 was correlated with the pathological grade and depth of invasion of patients with bladder urothelial carcinoma(p<0.05),and the lower expression level of IncRNA AC 104695.4 suggested that patients with bladder urothelial carcinoma have a deeper degree of infiltration and a higher pathological grade.5.The area under the ROC curve of IncRNA AC 104695.4 in diagnosing bladder urothelial carcinoma was 0.889(p<0.01),and it indicates that IncRNA AC 104695.4 has a certain accuracy in the diagnosing bladder urothelial carcinoma.In addition,the maximum value of the index was 0.66,the corresponding IncRNA AC 104695.4 value was 21.90,and its diagnostic sensitivity and specificity were 89.4%and 76.6%.Conclusions:1.LncRNA AC 104695.4 is lowly expressed in bladder urothelial carcinoma and bladder cancer cell lines.In addition,lncRNA AC104695.4 may also be lowly expressed in other urological cancer tumor tissues with pathological findings of urothelial carcinoma.2.LncRNA AC 104695.4 has a high positive detection rate and a certain accuracy in diagnosing bladder urothelial carcinoma,and the lower expression level of IncRNA AC 104695.4 suggests that patients with bladder urothelial carcinoma have a deeper degree of infiltration and a higher pathological grade.
