Objective: To observe the effects of Yanggan Yishui granule on blood pressure,urinary Microalbumin,renal fibrosis,plasma Ang?,AT1 R expression level,renal Ang?,AT1 R,TRPC6 protein in SHR rats with early renal injury,to explore the possible cellular and molecular mechanism of Yanggan Yishui granule in the treatment of hypertensive renal injury,and to expand its ideas and methods for the treatment of hypertensive renal injury.Methods:(1)Forty spontaneously hypertensive rats(SHR)were fed with high-fat diet for 4 weeks.They were randomly divided into SHR+Y group(Yanggan Yishui granule group),SHR+B group(benazepril group),SHR+Y+B group(Yanggan Yishui granule +benazepril group)and SHR group.Another 10 WKY rats matched by age and sex were selected.(2)The rats were given Yanggan Yishui granule 2.7g in SHR+Y group and benazepril0.9mg/kg/d in kg/d,SHR+B group according to benazepril at 10 o'clock in the morning,and Yanggan Yishui granule and benazepril were given to SHR+Y+B at 10 o'clock in the morning and 19 o'clock in the evening,respectively.Both SHR group and WKY group were given 2ml saline by intragastric administration at 8: 00 am.(3)The blood pressure of rats was measured by tail artery manometer before treatment and 4 weeks after treatment.(4)According to the method of kit use,the urine microalbumin of rats was detected before treatment and 4 weeks after treatment.(5)The contents of Ang? and AT1 R in serum were determined according to the usage of the kit.(6)The protein expression levels of Ang?,AT1 R and TRPC6 in kidney tissue of each group were measured by Western blot method.microscope.(8)The data were analyzed by SPSS 24.0 software.Results:(1)Systolic blood pressure of rats before and after treatment:Compared with WKY group,systolic blood pressure in SHR,SHR+Y,SHR+B and SHR+Y+B groups increased significantly,and compared with SHR group,after 4 weeks treatment,the systolic blood pressure in SHR+Y group,SHR+B group and SHR+Y+B group was significantly lower than that in SHR group,and there was significant difference between SHR+Y group,SHR+Y+B group and SHR+B group.SHR+Y group was higher than SHR+B group after treatment,and SHR+Y+B group was lower than western medicine group.(2)Urinary Microalbumin:Before treatment,SHR,SHR+Y,SHR+B and SHR+Y+B group were significantly higher than WKY group,and after treatment,SHR+Y,SHR+B and SHR+Y+B group were significantly lower than SHR group,SHR+Y group was higher than SHR+B group,SHR+Y+B group was lower than SHR+B group.(3)The content of Ang? and AT1 R in serum:The contents of serum Ang? and AT1 R in SHR,SHR+Y,SHR+B and SHR+Y+B groups were significantly higher than those in WKY,while the contents of serum Ang? and AT1 R in SHR+Y,SHR+B and SHR+Y+B groups were significantly lower than those in SHR group,and the contents of Ang? and AT1 R in SHR+Y group were higher than those in SHR+B group and SHR+Y+B group compared with SHR+B group,while those in SHR+Y+B group were lower than those in SHR+B group.(4)Protein expression of Ang?,AT1 R and TRPC6 in renal tissue:The expression of Ang?,AT1 R and TRPC6 in SHR group was significantly higher than that in WKY group,while the expression of Ang?,AT1 R and TRPC6 in SHR+Y,SHR+B and SHR+Y+B group was lower than that in SHR group,and the expression of Ang?,AT1 R and TRPC6 in SHR+Y group was higher than that in SHR+B group,while Ang?,AT1 R and TRPC6 in SHR+Y+B group was lower than that in SHR+B group.(7)The pathological changes of kidney were observed by HE staining and electron(5)In WKY group,the structure of renal tissue was intact,the size and shape of glomerular sac cavity was normal,the structure of glomerular capillary loop was clear,the structure of proximal renal tubule,medullary loop(thin segment)and distal renal tubule was not abnormal,the epithelial cells of renal tubule were neatly arranged,and the basement membrane was not thickened.In SHR group,renal glomerular sac cavity dilated,protein mucus exudation could be seen in the cyst cavity;capillary endothelial cells in glomeruli were necrotic,the number of capillary loops was rare;renal tubular epithelial cells were granular degeneration;a small amount of renal tubules could be seen protein tubule type.In SHR+B group,the structure of renal tissue was intact,the structure of glomerular capillary loop was clear,the size of glomerular sac was normal,the epithelial cells of proximal convoluted tubules were arranged regularly and orderly,the structure of brush margin was rich and normal,and protein tubules could be seen in individual tubules,and the epithelial cells of distal convoluted tubules showed granular degeneration and mild edema.In SHR+Y group,the structure of renal tissue was intact,the structure of glomerular capillary loop was clear,the size of glomerular sac was normal,some glomerular basement membrane was slightly thickened,the epithelial cells of proximal convoluted tubules were arranged regularly and the brush border was normal,and the epithelial cells of distal convoluted tubules were arranged regularly.In SHR+Y+B group,the structure of renal tissue was intact,the structure of glomerular capillary loop was clear,and the size of glomerular sac was normal;the epithelial cells of proximal convoluted tubules were arranged regularly and some of the brush edges of cells fell off slightly;in the visual field,the epithelial cells of distal convoluted tubules were slightly edematous and the cytoplasm was lightly stained.Conclusion:1.Yanggan Yishui granule can effectively reduce blood pressure and urinary Microalbumin excretion in SHR rats.2.Yanggan Yishui granule can reduce the content of Ang? and AT1 R in serum of SHR,reduce the protein expression level of Ang?,AT1 R and TRPC6 in kidney tissue,and reduce renal damage,which is beneficial to the improvement of renal damage in the early stage of hypertension.3.Yanggan Yishui granule can enhance the protective effect of benazepril on kidney,and the effect of combined use is better than that of benazepril alone and traditional Chinese medicine alone.
