Herba Houttuyniae,a kind of medicine edible plant ingredient,genus Saururaceae Houttrynia Cordata Thunb with the bioactive functions of anti-inflammatory,antioxidant,myocardial protection,antibacterial,antivirus,antitumor,as well as increase immunity etc.Therefore,the essential oil extracted from Herba Houttuyniae show a promising value in food functional factors and pharmaceutical fields.However,the protective effect of the essential oil on myocardial injury induced by doxorubicin(DOX)has rarely been reported.Thus,we focus on the essential oil extract from Houttuynia Houttuynia as the object of this study,analyzing its components by GC-MS.Besides,antioxidant and bacteriostatic tests of essential oil were carried out in vitro.Subsequently,the DOX myocardial injury model was established to study the protective mechanism of the essential oil on the myocardium.Finally,study analyzed the changes in urine metabolism of rats by using UPLC-Q-TOF/MS instrument after the intervention of essential oil,and results reveal that the essential oil possesses protective effects on myocardial injury.The main research contents are summarized as follows;(1)The composition analysis,antioxidant and bacteriostatic activities of the essential oil from Houttuynia Cordata were determined.The extraction rate of essential oil was 0.1097%by steam distillation.The main components of essential oil were determined by GC-MS and 28 compounds were identified.The scavenging effect of essential oil on DPPH free radical,the determination of reducing power,the antioxidant activity of metal ion chelating ability,the antibacterial activity of essential oil,the minimum inhibitory concentration(MIC)and the factors of antibacterial stability were investigated successively.The conclusion suggested that essential oil possessed antioxidant and antibacterial activity.(2)The study of the essential oil for its protective effect on the myocardial damage caused by doxorubicin.The experimental model of myocardial injury was established induced by DOX.The general situation,the whole blood routine test,the determination of LDH activity in heart tissue,the staining of heart tissue HE,and the test of the rats cecum content determination of short chain fatty acids(SCFAs)were measured respectively.Routine blood and SCFAs testing results revealed that DOX has an effect on platelets in the blood plasma,white blood cells and cecal contents SCFAs.After being given the essential oil,the platelets and SCFAs of rats being increased,while no significant difference,so that the essential oil haven't caused significant influence on bone marrow toxicity and intestinal toxicity induced by DOX.Nevertheless,LDH activity and HE staining results manifested that the essential oil possessed protective effects on myocardial injury.(3)The discussion on the protective mechanism of essential oil on myocardial injury mediated by doxorubicin.Results declared that the essential oil increase the liveness of SOD,CAT and GSH,decrease the content of MDA significantly,which can improve the antioxidant ability of rats.The liveness of TLR-4,NF-?B p65 and IL-1? were reduced according to inhibiting the NF-?B pathway to restrain inflammatory response.It mediates autophagy by increasing the expression of NF-?B p62 and decreasing the expression of LC3-?.Moreover,through reducing the apoptosis rate of myocardial cells and the caspase-2,caspase-3 and caspase-12 levels,and increasing the liveness expression of Hsp-70 to inhibit cell apoptosis.The results indicate that the essential oil has synergistic effects on the mechanism of anti-oxidative stress,inhibition of inflammatory response,regulation of autophagy and inhibition of apoptosis to protect cardiac tissue.(4)The research based on the metabolomics UPLC-Q-TOF/MS.The application of UPLC-Q-TOF/MS instrument to acquisition of rat urine metabolites,by using PCA and PLS-DA to screen differences metabolites,combining with the secondary metabolites mass spectrometry and database to identify differences,which showed that the identified 17 significant difference in urine,the identification of biomarkers of input MetaboAnalyst platform analysis of metabolic pathways by four metabolism,among which phenylalanine metabolism was the most significant one.
