Research On Sensibiligens In The Injection Of Heartleaf Houttuynia Herb (IHHH)

Objection:To screen out sensibiligens in the Injection of Heartleaf Houttuynia Herb(IHHH) by way of antibody reaction on antigen specifically.Method:The sera were collected to detect IgE concentration by using ELISA after Guinea pigs were sensitized by intradermal injection of IHHH. IgE is purified and isolated by salting out method and column chromatography to combine with sensibiligens in IHHH. After comparison of differences in chromatographic fingerprints between the original preparation and reacted residua for IHHH, according to the loss degrees of the characteristic peaks in chromatographic fingerprints, we could identify the sensibiligens of IHHH.Results:22 kinds of sensibiligens were screened out as follows: tetradecanoic acid; 7,9-Di-tert-butyl-1-oxaspiro(4,5)deca-6,9-diene-2,8-dione; 5-methyl-2-(1-methylethyl)-(1.α., 2.β., 5.α.)-(.+/-.)-cyclohexanol; 2,6,10-trimethyl-pentadecane,; heptadecane; 2,6,10,14-tetramethyl-pentadecane; dodecanoic acid; tetratriacontane; 4-methyl-1-(1-methyleth- yl)-3-cyclo hexen-1-ol; 2,6,10,14-tetramethyl-hexadecane;β.-myrcene; 2-(hexa decyloxy)-ethanol; 3-hexyldihydro-2(3H)-furanone; 2-methyl-hexadecane; heptane,6,6-dimethyl-2-methylen×10-, (1S)-bicyclo[3.1.1]; nonadecane; octadecanoic acid,2-hydroxyethyl ester; bornyl acetate; 2-undecanone; n-decanoic acid;α.,.α.4-trimethyl-; 1,6-Octadien-3-ol, 3,7-dimethyl-3-cyclohexene-1-methanol. Sensibiligens of metabolites are 4-Isopropyl-1,3-cyclohexanedione; Phenanthrene-9,10-dione,1-isop-ropyl-7-methyl-;.beta.,6.beta.-Epoxy-17-oxo-6.alpha.-pentyl-4-nor-3,5-se co androstan-3-oic acid, methyl ester; 1-Decanol,2-hexyl-; 7-Methyl-octadecane; Methoxyacetic acid,4-tridecyl ester; Dodecane, 3-cyclohexyl-; Acetamide,2-(1 H-purin-6-ylthio)-N-[(tetrahydro-2-furan-yl)methyl]-; 2-Hydroxymethyl-9-[.beta.-d-ribofuranosyl] hypoxanthine; Hexacontane; 6,7-Epoxy-trans-syn-tricyclo[7.3.0.0(2,6)] dodecane, (+-) (6S,7R)-; Naphtho[2,1-b]pyran-1-one,3-ethenyl-3,7,7-trimethyl-6,10,10 b-trihydroxy-5-[(3-isobutylamino-3-oxopropanoyl)oxy]-perhydro-;1R,3-tr ans-Dimethoxy-2-methylcyclohexane; 4,8-Dimethylheptacosane; Dotria-contane; Piperazine, 1-ethyl-4-(4-piperidyl)-; 4-Hydroxyiminomethyl-2, 2,5,5-tetramethyl-3-imidazoline-1-oyl; 5,8-Tridecadione; 8,11-Octadeca-diynoic acid, methyl ester; Hexanoic acid,3-(2,2,3,3-tetramethyl cyclopropylidene methylidene)-4-methyl-;Conclusions:Sensibiligens in IHHH were screened. These provide the basis of studying the clinical allergic reactions mechanism and excluding sensibiligens in IHHH, thus is expected to resume its clinical use.
