| Objective: By observing the liver color,daily behavior,and defecation of Functional Dyspepsia(FD)rats model with Liver Stagnation and Spleen Deficiency syndrome(LSSD),determining the gastrointestinal motility of FD rats model of LSSD,SCF content of abdominal aorta serum,expression of SCF/c-kit pathway in gastric antrum tissue,and the effects of interstitial cells of Cajal(ICCs)apoptosis in gastric antrum tissue,to study the mechanism of Shugan Hewei Recipe in treating FD with LSSD.And to provides theoretical basis for subsequent drug development and clinical use.Methods: Forty SPF male SD rats were randomly divided into normal control group(NC),model control group(MC),Mosha group(MS)and Shugan group(SH)after two days of adaptive breeding.(1)Modeling: Rats in the NC group were normally reared,and the other three groups were treated with the tail-clamp stimulation +irregular diet composite factor method to model the FD rat model of LSSD.The modeling period was 2 weeks.(2)Drug treatment: After the modeling was completed,it was the drug treatment period,lasting 2 weeks.During this period,the rats in the NC group and the MC group were given saline every day,ig.The MS group was given a solution of mosapride citrate suspension(0.15mg/ml),ig.And the SH group was given Shugan Hewei solution for treatment(concentration of crude drug 0.95g/ml),ig.The intragastric administration was 1ml/100 g.(3)Indicator detection: After the administration,the gastric emptying rate and the small intestine advancing rate of each group of rats were measured.The TUNEL method was used to determine the apoptosis of ICCs in the gastric antrum of each group of rats.Abdominal aorta serum SCF content,immunohistochemical method was used to detect the expression of c-kit,SCF in gastric antrum tissue of rats,and Western-blot method was used to detect the expression of c-kit,SCF protein in gastric antrum tissue of rats in each group.Results: 1.After the modeling,the model rats had withered hair,behavioral fatigue,and constipation.The weight of the MC,MS,SH group were significantly lower than the weight of the NC group(P<0.01).2.After the administration,the weight of the MC group still showed significantly difference compared with the NC group(P<0.01).And the weight between the NC,SH and the MS group showed no statistically significantdifference(P>0.05).3.The small intestinal advancing rate and the gastric emptying rate of the MC group rats showed significantly differences than those rate of the NC group(P<0.05).The gastric emptying rate and small intestinal advancing rate of the SH group and MS group were not significantly different from those of the NC group,the differences were statistically significant(P> 0.05).4.The TUNEL method showed that the apoptosis of ICCs in gastric antrum tissue of MC rats was significantly higher than that of NC rats,and there was no significant difference in the apoptosis rate of ICCs between NC group,MS group and SH group(P> 0.05).5.Elisa results showed that the serum SCF content of MC rats was significantly lower than that of NC group,MS group and SH group(P <0.05);the serum SCF content of MS group,SH group and NC group showed no significant difference(P> 0.05);Immunohistochemical results showed that the expressions of c-kit and SCF in gastric antrum of NC group,MS group and SH group were significantly higher than those of MC group(P <0.01).The c-kit and SCF protein expression in gastric antrum tissue of the MS group and SH group increased significantly compared with the MC group(P <0.01).Conclusions: 1.Based on the observation of animal macroscopic signs and body weight,it shows that Shugan Hewei Recipe can effectively relieve the symptoms of mental and behavioral discomfort in FD rats with LSSD.2.Through the detection of various indicators,Shugan Hewei Recipe can enhance the expression of c-kit and SCF in FD rats with LSSD,up-regulate the SCF/c-kit signal pathway,relieve ICCs apoptosis,and effectively improve gastrointestinal motility in FD rats of LSSD. |
