Objective:To study the regulatory mechanism of Wnt/?-catenin signaling pathway in angiogenesis after cerebral infarction,to observe the mechanism of scalp acupuncture combined with rehabilitation training to intervene in angiogenesis in mice after cerebral infarction,and to rebuild brain function after cerebral infarction and regeneration mechanism provide neurobiological clues;provide experimental basis for the study of acupuncture treatment of cerebral infarctionMethods:15 0 healthy male SD rats,280±20g,were randomly divided into sham operation group,model control group,scalp acupuncture group,exercise group,scalp acupuncture-exercise group,30 in each group.Each group was further divided into three subgroups of 7 days,14 days and 21 days by random number method,with 10 animals in each group.The animals were kept in cages by groups,and the rat food and water prepared by the animal center were uniformly fed,maintaining a normal 12-hour day and night cycle.The MCAO cerebral ischemia-reperfusion model was prepared by referring to the suture method reported by Zea-Longa.The sham operation group and the model control group received no treatment and were only grasped under the same conditions.In the treatment,the exercise group was given treadmill training treatment,while the scalp acupuncture plus exercise group was given treadmill training at the same time.At each time point,the rat's improved neurological deficit score,TTC to measure cerebral infarction volume,HE to observe brain tissue morphological changes,and immunohistochemical method to observe Wnt/?-Catenin signaling pathway related factors ?-catenin,cyclin D1,and growth factor-related gene VEGFResult:1.Rat behavior changes:Compared with the sham operation group,the rats in each group showed neurological deficit symptoms after modeling.After intervention treatment for 7d,14d and 21 d in each group,compared with the model group,the scores of the degree of neurological deficits in each treatment group were smaller than those in the model group,(P<0.05);The scores of the scalp acupuncture-exercise group at each time point were less than that of the pure The difference between the stab group and the exercise group(P<0.05)was statistically significant2.Morphological changes of the brain tissue:The morphology of the brain tissue of the sham operation group was normal,arranged in an orderly manner,and the vessel lumen was clear and full.At each time point in the model group,obvious ischemia-like changes in the brain tissue,sparse tissue arrangement,reduced cells,and condensed nuclei were seen.The morphology of the brain tissue of the treatment group was better than that of the model group at all time points and the improvement of the morphology of the scalp acupuncture-exercise group was better than that of the scalp acupuncture group and the exercise group,which was most obvious after 21 days of treatment3.Cerebral infarction volume rate test:No cerebral infarction was detected in the sham operation group,and the cerebral infarct volume rate of each treatment intervention group at each time point was smaller than that of the model group(P<0.05);The volume of cerebral infarction in the scalp acupuncture-exercise group was smaller than that of other treatment Group,(P<0.05),the difference was statistically significant4.Changes in Wnt/?-catenin signaling pathway angiogenesis-related proteins ?-catenin,cyclin D1,and VEGF:The number of positive proteins in the Wnt/?-Catenin signaling pathway angiogenesis-related protein model group and treatment groups increased,the number of positive proteins in each treatment group was greater than that in the model group,and the number of positive proteins in the scalp acupuncture+exercise group was greater than that in the scalp acupuncture group and the exercise group(P<0.05)Conclusion:1.Scalp acupuncture combined with exercise therapy can effectively improve the degree of neurological deficits in MCAO rats,promote pathological brain tissue repair,and reduce the volume of cerebral infarction2.Scalp acupuncture combined with exercise therapy can increase the expression level of angiogenesis-related factor VEGF3.The mechanism of scalp acupuncture combined with exercise therapy to promote angiogenesis in MCAO rats may be related to the expression of related factors ?-catenin and cyclin D1 on the Wnt/?-catenin signaling pathway.
