Objective:Hypertension is a chronic disease caused by genetic and environmental factors,which seriously threatens human health.Nowadays,association analysis based on SNPs of genes is an important method in genetic research of complex diseases.The purpose of this study was to investigate whether the three loci rs1801131,rs1801133 and rs9651118 of MTHFR gene and rs117353193 of TCN2gene are related to the risk of hypertension and the level of blood pressure in Han population in northeastern China.At the same time,it is also analyzed whether the interaction between genes and genes will affect the occurrence or development of the disease,and whether it will affect the level of blood pressure,which will help to elucidate the complex pathogenesis of hypertension and provide a theoretical basis for the prevention and treatment of hypertension.Methods:In this study,a total of 953 patients with hypertension and 1103 controls were enrolled from Shenyang,Fushun and Panjin City,Liaoning Province in northeast China.The epidemiological data and clinical indexes of the subjects were sorted out,the original data were integrated,and the related information resource database was preliminarily constructed.After the informed consent of the subjects,10ml of peripheral venous blood was taken and placed in the anticoagulant tube of EDTANa4.The blood indexes of all the subjects were detected by automatic biochemical analyzer.After DNA was extracted by human blood genomic DNA kit method,the concentration and purity of DNA were determined.After meeting the requirements of the follow-up test,it was stored in deep frozen refrigerator at-80?.Four gene loci SNPs were genotyped by TaqmanTMprobe real-time fluorescence quantitative PCR.The association between the gene loci and hypertension and its effect on blood pressure were analyzed by correlation statistical software.At the same time,the general multi-factor dimension reduction method was used to analyze the gene-gene interaction.Results:The results showed that there was no significant difference in the genotypes distribution and the alleles frequencies of rs1801131,rs1801133 and rs9651118 of MTHFR gene and rs117353193 of TCN2 gene between the case and control?P>0.05?.Binary regression analysis of environmental factors showed that waistline?P?0.006?and BMI?P?0.016?were significantly related to the occurrence of hypertension.In univariate logistic regression analysis,there were no significant effects of MTHFR rs1801131,rs1801133,rs9651118 and TCN2 rs117353193 on the occurrence of hypertension under additive,dominant and recessive genetic models.However,after further adjusting the important confounding factors such as sex,age,waistline,BMI,smoking frequency,FBG,TC and TG,the results showed that TCN2 rs117353193 had a protective effect on hypertension under the dominant genetic model?AA+GA vs GG?,and the risk of hypertension in AA+GA genotypic carriers was 0.56 times higher than that in GG genotypic carriers[OR=0.56,95%CI?0.32-0.99?,P?0.045].In addition,there were no significant differences in multiple linear regression analysis,comparative analysis of blood pressure,haplotype analysis of MTHFR gene and analysis of gene-gene interaction.Conclusion:In summary,our study suggests that the TCN2 rs117353193 gene polymorphism might serve as protective factor in hypertension,whereas the MTHFR rs1801131,rs1801133,and rs9651118 gene polymorphism may not be significantly correlated with the susceptibility of hypertension in Han population of Liaoning Province.
