Objective: To establish the pathological model of thoracic aorta in GK rats,observe and record the physiological indexes of GK rats,and explore the changes of oxidative stress indexes 8-iso-PGF2?,Ox-LDL and GSH-Px in GK rats under the intervention of Shenqi compound.Methods: 100 GK rats,male and 8 weeks old,were fed with high fat diet for 4weeks to simulate the establishment of diabetic macrovascular(thoracic aorta)disease model.The rats were randomly divided into four groups: model group(M group),Chinese medicine treatment group:high dose group(ZH Group),middle dose group(ZM group),low dose group(ZL group),western medicine treatment group(W Group).20 in each group.Another 20 normal Wistar rats of the same age and sex were used as the control group(Group C).The rats in the model group and the control group were given distilled water once a day.The rats in each group were killed at the end of 12 weeks.Observe the general situation of rats in each group,detect the weight,fasting blood glucose,serum8-iso-PGF2?,Ox-LDL and GSH-Px level,he staining,observe the pathological tissue of rat thoracic aorta through microscope,record the thickness of the wall of the great vessels,observe and record the apoptosis of the aortic tissue of GK rats in each group under TUNEL fluorescent staining,and calculate the apoptosis rate of the aortic tissue.result: Compared with the control group,the model group was in a state of continuous hyperglycemia with poor mental health and slow weight growth.HE staining was used to observe the pathological tissues,which showed that the vascular endothelium was damaged,the cells were swollen and fell off,TUNEL staining was used to observe,which showed that the number of apoptotic cells was significantly increased.Through the calculation of apoptotic rate,it was found that the apoptotic rate was increased The results showed that the oxidation index of 8-iso-PGF2?,Ox-LDL and GSH-Px were increased and decreased,which accorded with the characteristics of diabetic thoracic aortic disease.After the treatment of Shenqi compound,the mental state of rats recovered,the blood glucose decreased,the oxidation index 8-iso-PGF2?,Ox-LDL decreased,the antioxidant stress index GSH-Px increased,the apoptosis of thoracic aortic endothelial cells decreased,the injury gradually recovered,almost normal.Compared with the former,the apoptotic rate decreased obviously,indicating that Shenqi compound has the effect of repairing the damaged cells of aorta.Conclusion:1.GK rats can be fed with high-fat diet for diabetic thoracic aortic disease modeling.2.The curative effect of Shenqi compound on diabetic thoracic aortic disease is definite.3.Shenqi compound can reduce the body's oxidative stress response,improve the damage of large blood vessels and prevent and treat diabetes mellitus complicated with large blood vessels.
