Adiponectin(APN)is a collagen-like protein mainly derived from adipocytes,which belongs to the C1 q family of complement factors.It has significant effects on improving insulin sensitivity,increasing fuel oxidation,and reducing vascular inflammation.It plays an important role in promoting metabolism and improving the function of various organs.At present,there are few studies on the relationship between serum adiponectin and essential hypertension(EH).In this experiment,high resolution melting curve method(HRM)was used to explore the relationship between serum adiponectin gene methylation and EH,as well as the changes of serum APN concentration in patients with EH,so as to provide a new theoretical basis for the diagnosis and prevention of EH.The serum of healthy control group(30 cases,from the Department of Physical Examination,affiliated Hospital of Beihua University)and EH group(70 cases,from the Department of Cardiology,affiliated Hospital of Beihua University)were collected,and the concentration of APN in serum was detected by enzyme-linked immunosorbent assay(ELISA).At the same time,serum free nucleic acid was extracted by serum free DNA extraction kit and treated with sulfite for Real-Time Quantitative PCR.The methylated standard(100%)and unmethylated standard(0%)DNA were used to make the standard curve,and the methylation status of the promotor of APN gene in serum was determined according to the standard amplification curves.Statistical software analysis was used to determine the correlation between serum adiponectin and its gene methylation and essential hypertension.The concentration of serum APN in EH group was very low,which was statistically different from that of the healthy control group(P<0.05).In EH group,the concentration of serum APN was negatively correlated with systolic blood pressure(SBP)and diastolic blood pressure(DBP),with statistical differences(P<0.05).In the HRM test,the standard curves of methylation degree 0%,10%,25%,50%,100% were arranged from left to right.Compared with the control group,the methylation level of adiponectin gene promotor in EH group was elevated significantly(P<0.05).There was no significant correlation between serum APN concentrationand gene methylation level in patients with EH(P>0.05).There is a significant change in the concentration of serum APN in patients with EH,and the degree of gene methylation is significantly increased,suggesting that adiponectin gene may be involved in the pathogenesis of EH,which may be useful for predicting the occurrence of EH and finding new early screening indicators.
