Astragali Radix(Huangqi)can be divided into wild-simulated huangqi and cultivated huangqi according to the cultivation method.The growth period of wild-simulated huangqi is at least 6 years,the root grows downward,the length can reach more than 1 m,the growth period is longer and it is difficult to dig.The cultivation method of cultivated huangqi is to raise seedlings for one year,then transplanting horizontally,with short growth period and easy harvest.However,it is an urgent problem whether the effect of cultivated huangqi is comparable to that of wild-simulated huangqi.Herbal pair is the experience summary and essence of long term medical practice in the past dynasties.Aiming at the characteristics of certain syndromes,taking the corresponding treatment as the premise and combining the performance and function of the medicinal flavor,the 2 ingredients are selected and matched together,which is the smallest unit of prescription compatibility.Herbal pair is more simplified,which is more conducive to clarify the interaction between drugs and the mechanism of action on the body.Therefore,in this study,the herbal pair of “Astragalus Raidx-Salvia miltiorrhiza”(Huangqi-Danshen)and focal cerebral ischemia rat model were used to compare the efficacy difference of wild-simulated and cultivated huangqi.Objective: In order to compare the difference in the content of chemical components and efficacy of wild-simulated and cultivated huangqi based on its compatibility with danshen in the treatment of focal cerebral ischemia model rats,which to provide theoretical basis for the difference in clinical efficacy and application of wild-simulated and cultivated huangqi.Method:(1)By replicating the rat model of focal cerebral ischemia,we set up Huangqi-Danshen group with different doses and nimodipine group as the positive drug.The traditional pharmacodynamic indexes were the evaluationof nerve function damage,brain water content and TTC staining pathological results of brain tissue sections.The 1H-NMR metabonomics were used to analyze the serum metabolic profile of each group.The establishment of a systematic comparison model and the drug efficacy of each administration group were initially determined.(2)A focal cerebral ischemia rat model was used to compare the therapeutic effects of wild-simulated and cultivated huangqi on huangqi-danshen herbal pair.The traditional medicinal indicators of neurological damage evaluation and brain tissue TTC staining and the target tissue ischemic brain metabonomic analysis,systemic analysis of the drug effect callback effect of each administration group.(3)To compare the contents of 12 active components of wild-simulated and cultivated huangqi in the water extract of herbal pair “Huangqi-Danshen”.Firstly,a LC-MS/MS method for the determination of 12 components in huangqi was established.Through the experiments of precision,repeatability and stability,the established method was studied.Finally,the content of the chemical components in the water extract of huangqi-danshen was detected by the established method,so as to compare the differences of the chemical components in the water extract of wild-simulated and cultivated huangqi,providing the basis for the subsequent comparison of the differences in the efficacy of wild-simulated and cultivated huangqi.(4)The grey correlation analysis method was used to analyze the content of compounds and the effect of treating cerebral ischemia in wild-simulated and cultivated husngqi,and to find chemical constituents related to the differences in the efficacy of wild-simulated and cultivated husngqi.Results:(1)The model of focal cerebral ischemia rats and the preliminary efficacy of huangqi-danshen showed that the rats in the model group showed the classical symptoms including sluggish,piloerection,irritability and did not fully stretch the right forepaw.TTC staining results showed that the braintissue of the model group had significant ischemic sites,and the serum metabolic profile changed.After the administration of huangqi-danshen,the above phenomena all have different degrees of callback.It indicated that huangqi-danshen has protective effect on cerebral ischemia,and the efficacy of the high-dose group was better than that of the positive group.(2)In a study comparing the efficacy of wild-simulated and cultivated huangqi combined with danshen in the treatment of focal cerebral ischemia in rats,nerve function damage and TTC brain tissue staining results showed that the effect of cultivated huangqi-danshen on the treatment effect was better than wild-simulated huangqi.And metabolomics analysis was used to obtain10 brain metabolites related to the model.The number of differential metabolites between each group was analyzed,and the wild-simulated huangqi-danshen group was found that 10 kinds of metabolites can be callbacked significantly,and 9 different metabolites can be callbacked in the cultivated huangqi-danshen group.In the degree of reversal of differential metabolites,cultivated huangqi-danshen group was better than wild-simulated huangqi-danshen group.(3)In the determination of 12 chemical components in the water extract of wild-simulated and cultivated huangqi combined with danshen,a new LC-MS/MS method was developed to determine the precision,repeatability and stability of the 12 components in huangqi.The method with low detection and quantitation limits,and is applicable to detect.The contents of12 chemical components of wild-simulated and cultivated husngqi in the huangqi-danshen extract were detected,and the contents of the chemical components in the wild-simulated huangqi were higher than those of cultivated husngqi.It was found that the content of saponins in the water extract of wild-simulated huangqi was higher than that in the water extract of cultivated husngqi,which may be the main reason for the difference in the efficacy of the two.(4)Based on the grey correlation analysis between the content of chemical components and the effect in treating cerebral ischemia ofwild-simulated and cultivated huangqi,these compounds with a greater correlation degree are 1(7,2'-dihydroxy-3',4'-dimethoxyisoflavane-7-O-?-D-glucoside)? 3(Astragaloside II)?6(Formononetin)?10(Onion)?15(saponin / flavonoid).Conclusion: In this study,traditional medicinal efficacy indicators and metabonomics methods were used to compare the differences in chemical composition and efficacy of wild-simulated and cultivated husngqi,which based on the compatibility environment of huangqi-danshen.Firstly,the efficacy of huangqi-danshen water extract in the treatment of cerebral ischemia rats was proved.Secondly,by comparing the effects of wild-simulated and cultivated huangqi on the compatibility of the same compound environment,we found two the cultivation methods of huangqi are effective,and the lower cost of cultivated huangqi is more effective.Compared with the content of chemical components,it is found that the content of wild-simulated huangqi is higher than that of cultivated huangqi.And through grey correlation analysis,it was found that the ratio of saponin /flavonoid content was the highest,indicating that the ratio of saponin /flavonoid may be the reason why cultivated huangqi is better than wild-simulated huangqi.
