Objective:Observation of acupoint thread embedding combined with moxibustion on Ulcerative colitis(UC)model rat colon mucosal interleukin-6(Interleukin 6,IL-6),tyrosine kinase(Just another kinase,JAK)The effect of transcription factor signal and transcription activator of transcription 3(STAT3)expression was discussed from the perspective of IL-6 / JAK / STAT3 signaling pathway to explore the mechanism of action of acupoint embedding thread combined with moxibustion on UC.Methods : 32 SD rats,half male and half female,were randomly selected as the normal group,and the rest were replicated by TNBS /ethanol enema to replicate the UC model.The model was determined to be successful rats were divided into a model group,a thread embedding group,a moxibustion group,and a moxibustion plus embedding grou,with 6 rats in each group.For treatment,bilateral“Tianshu(ST 25)”,“Shangjuxu(ST 37)” and “Dachangshu(BL 25)” were selected.The moxibustion group performed mild moxibustion interventions for 10 min each time,once a day for a total of 14 interventions.The catgut embedding group received simple catgut embedding intervention at acupoints once a week,twice in total.The intervention method of thread embedding plus moxibustion group was the same as that of thread embedding group and moxibustion group.The thread embedding intervention was carried out 6 hours after mild moxibustion intervention.The general condition,body mass,defecation character and bleeding of rats in each group were observed,and the disease activity index(DAI)was evaluated.The histomorphology of colonic mucosa was observed by HE staining.interleukin-6(IL-6),tyrosine kinase(JAK),transcriptional activator 3(STAT3)were detected by immunohistochemistry and Western blotting respectively Expression.Results :1.General conditions:Normal group has dense hair and good mental state,drinking water,normal feeding,flexible movements,stool formation,no abnormal secretions around the anus,and the weight is increased compared with that before;in the model group,the hair is disordered and lusterless,the activity and spirit are poor,irritable,the back of the arch is piled up,the appetite is poor,the stool is thin or not formed,the anus is dirty,red or accompanied with pus secretion;in the three treatment groups,one is The general situation was improved in different degrees compared with the model group.2.Disease activity index score:Compared with the normal group,DAI in the model group was significantly increased(P <0.01);compared with the model group,DAI in the thread embedding plus moxibustion group was significantly lower(P < 0.01),DAI in the thread embedding plus moxibustion group and moxibustion group were significantly lower(P <0.05);DAI in the thread embedding plus moxibustion group was significantly lower than that in the thread embedding and moxibustion group(P < 0.01).3.HE staining microscopic examination showed that the colon tissue structure of the normal group was intact and clear,with a small amount of inflammatory cell infiltration,and no edema,hyperemia,erosion,ulcers;the model group had a severe defect of the upper layer of colonic mucosa,with superficial ulcer distribution and irregular glandular,glandular necrosis,atrophy,fuzzy structure,reduced mucus secretion,edema of the mucosal lamina propria and submucosa with a large number of inflammatory cell infiltration.In each treatment group,the colon tissue injury of the rats was improved to different degrees.Mucosal healing,mucosal epithelial intact or complete,mild edema of the mucosa and submucosa,infiltration of a small amount of inflammatory cells,the most significant improvement was in the embedding thread plus moxibustion group.4.Colonic mucosal IL-6 positive cell expression : IL-6 positive cells were expressed in the colonic mucosal tissue of rats in all groups,and the positive cells were yellow or brownish yellow.Compared with the normal group,the expression level of the model group increased significantly(P <0.01);compared with the model group,the expression level of each treatment group decreased significantly(P <0.01);the expression level of the embedding plus moxibustion group was significantly lower than that of theembedding group and Moxibustion group(P <0.01).5.Colonic mucosal JAK positive cell expression:JAK positive cells were expressed in the colonic mucosa of each group of rats,and the positive cells were yellow or brown-yellow.Compared with the normal group,the expression level of the model group increased significantly(P<0.01);compared with the model group,the expression level of each treatment group decreased significantly(P<0.01);the expression level of the embedding plus moxibustion group was significantly lower than that of the embedding group and Moxibustion group(P <0.01).6.Colonic mucosal STAT3 positive cell expression:STAT3 positive cells were found in the colonic mucosa of rats in each group,and the positive cells were yellow or brownish yellow.Compared with the normal group,the expression level of the model group increased significantly(P<0.01);compared with the model group,the expression of each treatment group decreased significantly(P<0.01);the expression level of the embedding plus moxibustion group was significantly lower than that of the embedding group and moxibustion group(P<0.01).7.IL-6 protein expression in the intestinal mucosa:Compared with the normal group,the expression level of the model group was significantly increased(P<0.01);compared with the model group,the expression level of each treatment group was significantly reduced(P<0.05,P<0.01);the embedding plus moxibustion group was significantly lower than the embedding group and moxibustion group(P<0.01).8.JAK protein expression in intestinal mucosa : Compared with the normal group,the expression level of the model group was significantly increased(P<0.01);compared with the model group,the expression level of each treatment group was significantly reduced(P<0.01);the embedding plus moxibustion group was significantly lower than the embedding group and moxibustion group(P<0.05,P<0.01).9.STAT3 protein expression in intestinal mucosa:Compared with the normal group,the expression level of the model group was significantly increased(P<0.01);compared with the model group,the expression level of the embedding plus moxibustion group and the embedding group was significantly reduced(P<0.01);and the expression level of the moxibustion group was lower than the model group,but the difference was not significant(P>0.05);the embedding plus moxibustion group was significantly lowerthan the embedding group and moxibustion group(P<0.01).Conclusion : Thread embedding plus moxibustion can reduce the disease activity index and improve the pathological damage of colonic mucosa in UC rats,and the effect is better than simple embedding thread and simple moxibustion.The mechanism may be through down-regulation of IL-6 / JAK / STAT3 signaling pathway-related factor protein content and inhibition of inflammatory injury.
