Objective:To investigate the therapeutic effect of acupoint catgut embedding on UC rats under the condition of deficiency and stasis,and to explore the effect of acupoint catgut embedding on the expression of Occludin and ZO-1 protein and m RNA from the perspective of intestinal mucosal mechanical barrier,and to explore the mechanism of acupoint catgut embedding in the treatment of UC.Methods: Forty-six SPF SD rats,aged 2-3 months and weighed 200±20g,were adaptively fed for one week.One week later,they were randomly divided into blank group,model group,embedding group and SASP group.The blank group was 10 rats,and the other three groups were 12 rats in each group.The animal model of UC disease-syndrome combination was prepared by intragastric administration of senna and adenine instages combined with external intervention,followed by enema with 2,4,6-trinitrobenzene sulfonic acid and 50% ethanol.After modeling,the concentrations of serum interl eukin6(IL-6),interleukin8(IL-8),tumor necrosis factor-?(TNF-?),D-xylose,free triiodothyronine(FT3),free thyroxine(FT4),cortisol(COR),and testosterone(T)were detected,and HE staining was performed to verify the modeling.Interventions began 10 days after successful modeling.The rats in the catgut embedding group were treated with catgut embedding at acupoints of “Zu sanli” “Tian shu”“Shen shu”“Pishu”“Da changshu” and“Ge shu”for 3 times,once every 14 days,for 6 weeks.SASP group was given intragastric administration of SASP once a day for 6 weeks.The general signs of rats,the disease activity index(DAI)score of rats,the general morphology of colon tissue and the degree of colonic mucosal injury were observed.The pathological changes of colon tissue were observed under optical microscope.Colonic mucosal damage index(CMDI)and tissue damage index(TDI)were scored.The expression levels of Occludin and ZO-1 were detected by RT-PCR and Western blot.the mental state,body weight,diet,hair,feces and other aspects,the rats in the syndrome model group showed perianal filth,paste stool,and thin stool.Some rats showed mucous purulent blood stool,crouching and lazy movement,squinting at the bow and back,sparse and glossless hair,significantly reduced food intake and water intake,and significantly reduced body weight.DAI score after treatment: after treatment,compared with the model group,the DAI scores of SASP group and embedding group were decreased,P<0.05;Compared with the SASP group,there was no significant difference in the embedding group,P>0.05.2.CMDI score and TDI score: compared with the blank group,the CMDI score and TDI score of the other three groups were significantly increased,P<0.05;Compared with the model group,the CMDI score and TDI score of the buried wire group and SASP group were decreased,P<0.05;Compared with the SASP group,the CMDI and TDI scores of the buried wire group were not significantly different,P>0.05;3.The expression levels of Occludin and ZO-1 protein in model group,SASP group,and ZO-1 group were significantly lower than those in blank group,P < 0.05,the expression levels of Occludin and ZO-1 protein in buried wire group were not significantly different,p > 0.05,compared with model group,the expression levels of Occludin and ZO-1 protein in embedding group and SASP group were increased,P < 0.05;compared with SASP group,the expression levels of Occludin protein in embedding group had no significant difference,p > 0.05,the expression level of ZO-1 protein in embedding group was increased,p <0.05;4.Occludin and ZO-1 gene transcription levels:Compared with the blank group,the m RNA expression levels of Occludin and ZO-1 in the other three groups were significantly decreased,P<0.05;After treatment,compared with the model group,the expression levels of Occludin and ZO-1m RNA in catgut embedding group and SASP group were increased,P<0.05;Compared with SASP group,there was no significant difference in the expression levels of Occludin and ZO-1 m RNA in catgut embedding group(P>0.05).5.Pathological results of colon tissue HE staining:The structure of colon tissue in blank group was intact and clear,no congestion andResults:1.Macroscopic characterization observation:Compared with edema,no ulcer,no necrosis and shedding.Compared with the normal group,the colon tissue of rats in the model group had obvious ulcers,mucosal epithelial cells fell off,gland structure was disordered,and a large number of inflammatory cells were seen in the mucosa and submucosa.Compared with the model group,the colon tissue congestion degree,gland damage degree and inflammatory cell infiltration degree in the catgut embedding group and SASP group were improved to varying degrees.Conclusion:1.Acupoint catgut embedding significantly improved the general conditions of UC rats,such as spirit,hair,weight,diet and feces.2.Acupoint catgut embedding has obvious effect on alleviating disease activity index(DAI)and colonic mucosal injury index(CMDI)score of UC rats.3.Acupoint catgut embedding can improve symptoms,promote ulcer healing,and effectively treat UC in rats with deficiency and stasis Its mechanism may be related to up-regulating the expression levels of Occludin,ZO-1 protein and Occludin and ZO-1 m RNA,and promoting the regeneration and repair of intestinal mucosal mechanical barrier in UC rats by maintaining the integrity and stability of intestinal epithelial TJ.
